摘要
目的比较高效液相色谱串联质谱法(HPLC-MS/MS)与电化学发光免疫分析法(ECLI)测定人血清中25-羟基维生素D浓度结果的相关性。方法收集66例需测定25-羟基维生素D浓度的血清样本,用HPLC-MS/MS法和ECLI法分别测定相同临床样本的25-羟基维生素D浓度。对测定结果进行统计分析和比对,评价两种方法测定结果的相关性。结果 66例样本中25-羟基维生素D浓度分布为4.89~103.50μg·L^(-1),HPLC-MS/MS法与ECLI法对25-羟基维生素D浓度测定的结果平均值分别为(26.91±14.17)和(39.13±22.66)μg·L^(-1),HPLC-MS/MS法测定浓度/ECLI法测定浓度比值为(148.37±31.68)%;2组测定结果方程式为yHPLC-MS/MS=1.48xECLI-0.78(r=0.926 8),相关性较好。2种方法测定浓度结果比较,差异有统计学意义(P<0.01)。结论 HPLC-MS/MS与ECLI测定人血清中25-羟基维生素D浓度的结果相关性较好,可通过方程式进行结果互相推算。
Objective To analyze the correlation between high performance liquid chromatography-tandem mass spectrometry( HPLC-MS/MS) and electro-chemiluminescence immunoassay( ECLI) for the determination of 25-hydroxy-vitamin D concentration in human serum.Methods The concentrations of 25-hydroxy-vitamin D in collected66 serum samples were determined by HPLC-MS/MS and ECLI,respectively. The correlation between the concentrations obtained by different methods was analyzed and evaluated. Results The concentration of 25-hydroxyl vitamin D in 66 samples was in the range of4. 89-103. 50 μg · L-1. The average concentrations of 25-hydroxy vitamin D by HPLC-MS/MS and ECLI method were( 26. 91 ± 14. 17)and( 39. 13 ± 22. 66) μg · L-1. The ratio of CHPLC-MS/MS/CECLIwas( 148. 37 ± 31. 68) %. The result equation was yHPLC-MS/MS= 1. 48 xECLI-0. 78( r = 0. 926 8). The P value evaluated by paired-samples t test showed that there was statistical difference between two methods( P〈0. 01). Conclusion There was good correlation between HPLC-MS/MS and ECLI for the determination of 25-hydroxy-vitaminD in serum. The results can be calculated through the equation.
