海人酸诱导的颞叶癫痫小鼠慢性期行为学变化

摘要 目的:研究海人酸(Kainic acid)所致的颞叶癫痫小鼠慢性期行为学改变及意义。方法:用海人酸构建癫痫小鼠模型作为实验组,对照组用生理盐水处理,2个月后分别进行Morris水迷宫实验、条件恐惧实验、高架十字实验和旷场实验。结果:Morris水迷宫实验中,后3d实验组小鼠的定位航行潜伏期长于对照组(P<0.01),实验组目标象限滞留时间短于对照组(P<0.01);条件恐惧实验中,场景联系性记忆实验实验组小鼠僵直时间百分比小于对照组(P<0.01),声音联系性记忆实验实验组小鼠僵直时间百分比小于对照组(P<0.01);旷场实验中,实验组小鼠活动路程长于对照组(P<0.01),实验组小鼠中间区域滞留时间短于对照组(P<0.01),实验组小鼠大小便次数比对照组多(P<0.01);高架十字实验中,实验组和对照组小鼠进入开臂的次数比无显著差别(P>0.05),两组小鼠在开臂的活动时间比较也无差异(P>0.05),但实验组小鼠进入两种臂的总次数较对照组小鼠多(P<0.05)。结论:海人酸诱导的颞叶癫痫小鼠慢性期有认知功能和情绪方面的障碍。 Objective:To research the chronic behavioral changes and significance in temporal lobe epileptic mice induced by kainic acid.Methods:Epileptic mice model was constructed with kainic acid as the experimental group,the control group was handled with physiological saline, nevertheless we have mice in a control group injected normal saline.After two month,the Morris water maze experiment, the condition fear experiment, the elevated cross experiment and the open field experiment were carried out in the two groups. Re- suits. In the Morris water maze experiment, the search latency period in the last three days of experimental group was longer than that of control group (P〈0.01).The retention time in the target quadrant in the experimental group is shorter than that in the control group (P〈0.01).In the condition fear experiment,the percentage of stiff time of the mice in the scene linked memory experiment of experimental group was less than that of control group (P〈0.01).The percentage of stiff time in the sound linked memory experiment of experimental group was less than that of control group (P 〈0.01).In the open field test, the distance of movement of mice in the experimental group was longer than that of the control group (P〈0.01),the retention time in the middle area of the experimental group was shorter than that of the control group (P〈0.01), the number of stool times in the experimental group is more than that in the control group (P〈0.01).In the elevated cross experiment, there was no significant difference in the number of open arms between the experimental group and the control group (P〉0.05),there was no difference in the time of open arm between the two groups (P〉0.05),but the total number of open arms in the experimental group was more than that in the control group (P〈0.05). Conclusion:The cognitive function and emotional behaviors are affected in the mice induced by Kainic acid with chronic attack.
