摘要 目的检测FOXF1蛋白在胃癌中的表达并探讨其表达差异与临床资料及预后的关系。方法收集160例胃癌患者,利用Oncomine基因芯片数据库预测FOXF1在胃癌中的表达,采用Western blot等方法检测胃癌及其癌旁正常组织中FOXF1蛋白的表达量,将胃癌中FOXF1表达低于癌旁组织的病例设为低表达组,其余病例设为高表达组,分析其与临床资料及病理特征的关系;应用Kaplan-Meier法分析FOXF1蛋白的表达差异与临床预后的相关性。结果 Oncomine数据库及Western blot实验均显示FOXF1蛋白在胃癌中相对表达量明显低于癌旁正常组织(P<0.05);FOXF1蛋白低表达与肿瘤的大小、分化及肿瘤分期密切相关(P<0.05)。生存分析显示FOXF1蛋白低表达组生存率明显低于高表达组(P<0.05)。结论 FOXF1蛋白参与了胃癌的发生和发展,可作为评估胃癌预后的指标。 Objective To detect the expression of FOXF1 protein in gastric cancer and to investigate the correla- tion between its expression and the clinical data or prognosis of gastric cancer patients. Methods The expression of FOXF1 protein in 160 gastric cancer patients and the adjacent normal tissues were predicted by Oncomine microarray data- base. The expression of FOXF1 protein was detected by western blot. The patients were divided into high expression group and low expression group according to the median value of FOXF1 expression. The clinical data of two groups were com- pared. Kaplan -Meier methods were used to analysis the correlation between FOXF1 protein and survival. Results On- comine database and Western blot experiments showed that the expression of FOXF1 protein in gastric cancer was signifi- cantly lower than that in normal tissues ( P 〈 0. 05 ). The expression of FOXF1 protein was closely correlated with tumor size, differentiation and tumor staging ( P 〈 0. 05 ). Survival analysis showed that the survival rate of low expression group was significantly lower than that of high expression group ( P 〈 0. 05 ). Conclusion FOXF1 protein is involved in the de- velopment and progression of gastric cancer, and may be helpful in evaluating the prognosis.
