芒果苷对自发性高血压大鼠交感神经形态学及MCP-1/CCR2信号通路的影响 被引量:6

Effects of Mangiferin on Sympathetic Nerve Morphology and MCP-1/CCR2 Signal Pathway in Spontaneously Hypertensive Rats

摘要 目的 :通过观察芒果苷对自发性高血压大鼠交感神经形态学及单核细胞趋化蛋白-1(MCP-1)及其受体CCR2表达量的影响,探讨自发性高血压大鼠是否存在交感神经炎症损伤以及芒果苷可能的抗炎药效及机制。方法 :以8只10周龄正常血压对照大鼠(WKY)作为空白组,以40只同龄SHR大鼠作为研究对象,随机分为模型组、苯那普利组、芒果苷高剂量组、芒果苷中剂量组和芒果苷低剂量组。各组动物按每天给药10 m L/(kg·d)连续灌胃,灌胃给药2个月后停药,杀鼠前禁食12 h。结果 :各组大鼠交感神经均未见明显病理形态学改变;免疫组化检测结果显示:与空白对照组比较,模型组交感神经MCP-1、CCR2水平显著升高(P<0.01);与模型组比较,各组给药大鼠交感神经组织中MCP-1、CCR2含量均有不同程度下降(P<0.05,P<0.01)。结论 :自发性高血压大鼠存在交感神经炎症损伤。芒果苷对自发性高血压大鼠交感神经组织病理形态学无显著影响,但可下调大鼠交感神经组织异常升高的炎症因子MCP-1、CCR2水平,对交感神经炎症损伤具有一定的保护作用。 Objective :By observing the mangiferin on sympathetic nerve morphology in spontaneously hypertensive rats and the expression of monocyte chemoattractant protein-1(MCP-1)and its receptor CCR2,to investigate the existence of spontaneously hypertensive rats and mechanism of anti-inflammatory effect of sympathetic nerve injury and inflammation of mangiferin may.Methods :By using 8 normal blood pressure at 10 weeks of age WKY rats served as normal controls.40 spontaneously hypertensive rats(SHR)were randomly divided into model group,benazepril group,the mangiferin-high dose group,the mangiferin middle dose group,the mangiferin low dose group.Animals in each group were given 10 m L/(kg·d)for days,and the rats were fed for a period of 2 months.Results :There were no obvious pathological changes in the sympathetic nerves of the rats in each group,the results of immunohistochemistry showed that :compared with the blank control group,the levels of MCP-1 and CCR2 were significantly increased in the model group(P〈0.01);compared with the model group,the contents of MCP-1 and CCR2 were decreased in different groups(P〈0.05,P〈0.01).Conclusion :There are sympathetic nerve injury in spontaneously hypertensive rats.Mangiferin has no significant effect on sympathetic nerve tissue and pathological morphology of spontaneously hypertensive rats,but can decrease the rat sympathetic nerve tissue abnormal elevation of inflammatory cytokines MCP-1 and CCR2,has a protective effect on sympathetic nerve injury.
