摘要
目的 :观察解毒化瘀生津方对干燥综合征小鼠颌下腺干扰素-γ(Interferon-γ,IFN-γ)、白介素-4(Interleukin-4,IL-4)蛋白含量表达的影响,探讨其作用机制。方法 :采用免疫诱导法建立干燥综合征小鼠动物模型,以解毒化瘀生津方灌胃治疗,并以硫酸羟氯喹片(Hydroxychloroquine Sulfate,HCQ)灌胃作为对照,采用酶联免疫吸附法检测颌下腺IFN-γ、IL-4蛋白含量表达,并对小鼠颌下腺IFN-γ、IL-4进行免疫组织化学法检测及免疫荧光染色观察。结果 :酶联免疫吸附法检测发现,模型组颌下腺IFN-γ、IL-4的蛋白含量及IFN-γ/IL-4比值高于正常组(P<0.05,P<0.01),中药中、高剂量组和硫酸羟氯喹组IFN-γ、IL-4蛋白含量及IFN-γ/IL-4比值低于中药低剂量组(P<0.05)。免疫组化方法检测小鼠颌下腺IFN-γ、IL-4的平均吸光度,模型组颌下腺IFN-γ、IL-4的吸光度高于正常组(P<0.05),中药中、高剂量组和硫酸羟氯喹组吸光度低于中药低剂量组(P<0.05),接近于正常组。免疫荧光技术观察,模型组荧光染色最强,正常组染色最弱。结论 :解毒化瘀生津方能降低干燥综合征模型小鼠颌下腺IFN-γ、IL-4蛋白含量的表达,恢复了Th1/Th2的平衡稳态,减轻颌下腺的病理损伤,与硫酸羟氯喹的作用相当。
Objective :To observe the protein content change of interferon-γ(IFN-γ)and interleukin-4(IL-4)in the submandibular glands of model mice with Sj?gren's syndrome,feeding all the mice with Jiedu Huayu Shengjin Decoction,to explore the its mechanism.Methods :To establish the mice model of Sj?gren's syndrome by immune induction,then feed the mice with Jiedu Huayu ShengjinDecoction,and feed the control group mice with hydroxychloroquine sulfate at the same time,finally detect quantity change of(interferon-γ)IFN-γ and(interleukin-4)IL-4 in the submandibular glands of model mice,which detected by immunohistochemistry and immunofluorescence staining,to observe the result.Results :Results by ELISA showed that the content of FIN-γ and IL-4 of the submandibular glands and the ratio of them in the model group was higher than the normal group(P〈0.05,P〈0.01),medium and high dose of Chinese medicine group and hydroxychloroquine sulfate group was lower than the low dose of Chinese medicine group(P〈0.05);About mean absorbance of IFN-γ and IL-4 in the submandibular glands by IHCA,the model group was higher than the normal group(P〈0.05);medium and high dose of Chinese medicine group and hydroxychloroquine sulfate group is lower than the low dose of Chinese medicine group(P〈0.05),which close to the normal group.Results by immunofluorescence technique showed that the fluorescence staining of model group was strongest,the normal group was the weakest.Conclusion :Jiedu Huayu Shengjin Decoction can reduce the level of IFN-γ and IL-4 in the mice submandibular gland,restore the balance homeostasis of Th1 and Th2,alleviate the pathological damage of submandibular gland,the effect of which is equivalent to hydroxychloroquine sulfate.
