藏药十八味党参丸抑制CIA大鼠滑膜组织炎性因子表达和细胞凋亡诱导 被引量:4

摘要 目的通过研究藏药十八味党参丸(TEP)对CIA大鼠滑膜组织中炎性因子表达的影响,探讨对胶原诱导性关节炎的疗效及其分子作用机制。方法将雌性SD大鼠随机分为空白对照组(Control)、模型组(CIA)、雷公藤组(TP)、TEP低剂量组(TEPⅠ)、TEP中剂量组(TEPⅡ)和TEP高剂量组(TEPⅢ),每组10只。CIA模型采用卡介苗溶液及牛胶原混合免疫法建立,各组大鼠通过药物灌胃6周后,取关节滑膜组织,采用HE染色法检测滑膜组织炎症程度;ELISA法检测各组大鼠滑膜组织白细胞介素-17因子的表达水平;免疫组化法检测各组大鼠滑膜组织肿瘤坏死因子-α的表达水平;Western blot法检测各组大鼠滑膜组织半胱天冬酶-3、核转录因子kappa B、B细胞淋巴瘤-2与相关X蛋白表达水平。结果与对照组相比,CIA模型组大鼠滑膜组织有明显的炎症反应,IL-17、NF-κB和TNF-α的表达明显增加(P<0.05),caspase-3表达显著降低(P<0.05),BCL-2/BAX比值显著增高(P<0.05)。与CIA模型组大鼠相比,TP组和TEPⅢ组大鼠滑膜组织炎症反应显著减轻,IL-17、NF-κB和TNF-α表达明显降低(P<0.05),caspase-3表达显著增加(P<0.05),BCL-2/BAX比值明显降低(P<0.05)。结论 TEP通过上调CIA大鼠滑膜组织中caspase 3表达,下调IL-17、NF-κB及BCL-2/BAX比值,可改善CIA大鼠滑膜组织的炎症反应。 Objective Rheumatoid arthritis is a common clinical aut0immune disease.The purpose of this study is to investigate the effect and molecular mechanism of Tibetan eighteen flavor dangshen pills( TEP )on collagen-in- duced arthritis in rats.Methods SD rats were randomly divided into control group, model group (CIA) , Tripterygium -treated group ( TP ), low-dose TEP treatment ( TEP I ), middle-dose TEP treatment group ( TEP II ) and high- dose TEP treatment(TEP m).cIA rat model was established by immunization with BCG solution and bovine colla- gen.Rats in each administration group were given intragastric administration for 6 weeks.The expression of IL-17 in the synovial membrane of rats was detected by ELISA.The expressions of caspase 3,NF-KB, BCL-2 and BAX ex- pression in synovial membrane were detected by Western blot.Results Compared with the control group, the expres- sion of IL-17 and TNF-α,the level of NF-KB ,and the ratio of BCL-2 to BAX in synovial membrane was signifi- cantly increased in the CIA model group( P〈0.05), while the expression of caspase 3 was significantly decreased (P〈0.05) .Compared with model group, the expression of IL-17 and TNF-α was significantly decreased in TP group and TEP III group(P〈0.05), NF-KB level, BCL-2 And BAX significantly decreased, caspase 3 expression was significantly increased(P〈0.05).Conclusion TEP can improve the synovial inflammation of CIA rats by in- creasing the expression of caspase 3 in CIA rats and decreasing the levels of IL-17, NF-KB ,TNF-α and the ratio of BCL-2 and BAX.
