灸“赫依穴”对慢性应激抑郁模型大鼠行为学及海马5-羟色胺的影响 被引量:3

Effect on Behavior and Hippocampus Serotonin(5-HT) of Chronic Stress Depression Rats by Moxibustion on "Heyi" Acupuncture Point

摘要 目的:观察灸"赫依穴"对慢性应激抑郁模型大鼠行为学及海马5-羟色胺(5-HT)含量的影响,探讨蒙医灸疗法的抗抑郁作用机制。方法:将雄性Wistar大鼠随机分为正常组、模型组、氟西汀组、灸赫依穴组,每组8只。采用慢性不可预知温和应激方法和孤养相结合的方法制备抑郁大鼠模型。采用体质量、旷场实验评价大鼠行为学变化,用ELISA方法检测大鼠海马5-HT的含量。结果:模型组大鼠体质量、旷场实验的水平运动和垂直运动次数、海马5-HT含量均明显低于正常组(P<0.01)。氟西汀组和赫依穴组大鼠体质量、旷场实验平行运动和垂直运动次数、海马5-HT含量均高于模型组(P<0.05)。氟西汀组和赫依穴组比较,两组大鼠体质量、旷场实验平行运动次数和垂直运动次数、海马5-HT含量比较,差异无统计学意义(P>0.05)。结论:灸"赫依穴"可改善慢性应激抑郁模型大鼠行为学变化,提高海马5-HT含量,具有明显的抗抑郁作用。 Objective: By observing the behavioral and hippocampus serotonin (5-HT) content changes of chronic stress depression model rats, this study aims to explore the Mongolian medicine moxibustion therapy's antidepressant mechanism. Methods: The male Wistar rats were randomly divided into normal group, model group, the fluoxetine group and "Heyi" group with 8 rats in each group. Combining of chronic unpredictable mild stress method and solitary keeping method, the depression model rats was prepared. The behavioral change was evaluated in rats according to the weight and field test. The contents of 5-HT was detected in rat's hip- pocampus by ELISA method. Results: Model group rats showed lower weight, horizontal movement, vertical movement of field test and 5-HT content in hippocampus than normal group (P〈0.01). The fluoxetine group and "Heyi" group rats showed higher weight, horizontal movement, vertical movement of field test and 5-HT con- tents of hippocampus than the model group (P〈0.05). Between the fluoxetine group and "Heyi" group, there was no significant difference in rat's body weight, horizontal movement and vertical movement of field test and the contents of 5-HT in hippocampus (P〉0.05). Conclusion: Moxibustion on "Heyi" acupuncture point can improve chronic stress depression model rat's behavior changes, and improve the content of 5-HT in hippocampus; it has obvious antidepressant effect.
