苦瓜总皂苷对阿霉素肾病大鼠P-GSK-3β表达的影响

摘要 目的分析苦瓜总皂苷对阿霉素肾病大鼠磷酸化糖原合成酶激酶-3β(P-GSK-3β)表达的影响。方法50只SD大鼠随机分为空白组、模型组、苦瓜总皂苷低、中、高治疗组,造模成功后给予8周治疗后处死大鼠,观察肾脏病理学,肾功能及P-GSK-3β的表达。结果与空白组相比,模型组12 h蛋白定量、BUN、Scr、TG和CHO增加(P<0.05),苦瓜总皂苷治疗组相较于模型组下降(P<0.05);与空白组相比,模型组TP、ALB、Bax和Bcl-2降低(P<0.05),苦瓜总皂苷治疗组相较于模型组升高(P<0.05);组织病理学检测,空白组肾脏无异常,模型组上皮细胞明显变性,苦瓜总皂苷治疗后明显改善;与空白组相比,模型组P-GSK-3β大量表达(P<0.05),苦瓜总皂苷治疗组相较于模型组明显降低(P<0.05)。结论苦瓜总皂苷可改善阿霉素肾病大鼠肾功能,与其可降低肾脏P-GSK-3β表达有关。 Objective To study the effect of momordicosides to P-GSK-3β in rats with adriamycin nephropathy.Methods 50 SD rats were randomly divided into blank group, model group, low, middle and high momordicosides dose treatment group, after built model, renal pathology, renal function and P-GSK-3β were detected.Results Compared with the blank group, the quantitative of 12 h protein, BUN, Scr, TG and CHO in the model group were significantly higher(P〈0.05), above mentioned indicators were lowed in momordicosides group compared with the model group(P〈0.05). Compared with the blank group, the levels of TP, ALB, Bax, and Bcl-2 were significantly low(P〈0.05), compared with the model group, they were higher in momordicosides treatment group(P〈0.05). Histopathological can be seen, there is no abnormality in the blank group, the epithelial cells were obviously denatured in the model group, after using momordicosides, it was significantly improved. Compared with the blank group, the expression of P-GSK-3β was significantly lower in the model group than in the model group(P〈0.05), the momordicosides was significantly improved compared with the model group(P〈0.05).Conclusion Momordicosides can improve the renal function of rats with adriamycin nephropathy, which may be related with it can decrease the expression of P-GSK-3β.
