摘要 目的:优化黄芪多糖标准提取物的醇沉工艺条件。方法:在单因素实验的基础上,以多糖含量、湿多糖得率的总评归一值(OD值)为指标,依据响应面设计原理,并运用Design Expert 8.0.5b软件中的BoxBehnken试验设计建立数学模型,考察醇沉浓度、醇沉时间、醇沉次数对醇沉工艺的影响。结果:最佳醇沉工艺条件为:醇沉浓度85%,醇沉时间13 h,醇沉次数2次,在此条件下验证的总评值为0.875 1,与预测值(0.900 4)偏差较小。结论:采用响应面法优化得到的醇沉工艺稳定、合理、可行,为黄芪多糖标准提取物的醇沉工艺提供依据。 Objective: To optimize the extraction conditions of Astragalus polysaccharides standard. Methods: The test was carried out on the basis of the single factor experiment and response surface design principle,the polysaccharide content and the yield value of wet polysaccharide(OD)were used as indexes,and the mathematical model of Box-Behnken was used in Design Expert8.0.5 b software. The effects of alcohol concentration,alcohol precipitation time and alcohol precipitation times on the alcohol precipitation process were investigated. Results: The optimum conditions of alcohol precipitation were as follows: alcohol concentration was 85%,alcohol precipitation time was 13 h and alcohol precipitation for twice. The total evaluation value was0.875 1,which was less than the predicted value(0.900 4). Conclusion: The process of alcohol precipitation was optimized by response surface methodology(RSM),which provided the basis for the alcohol precipitation process of the standard extract of Astragalus polysaccharides.
