miR-708与JAK1在大鼠脑缺血再灌注损伤中的表达及调控关系 被引量:1

摘要 目的初步探讨miR-708在大鼠脑缺血再灌注损伤中的作用及机制。方法选取25只雄性SD大鼠,随机抽取10只作为假手术组,其余大鼠采用线栓法构建大鼠脑缺血再灌注模型(MACO),将术后存活的10只SD大鼠设为缺血再灌注1d组,采用实时定量荧光PCR法(real time,PCR)比较各组大鼠外周血和脑组织的miR-708表达及脑组织中JAK1 mRNA的表达;蛋白免疫印迹法(Western blot)检测各组大鼠脑组织中JAK1蛋白的表达水平。结果与假手术组相比,实验组外周血及脑组织中miR-708mRNA表达显著降低(P<0.01),而脑组织中JAK1 mRNA表达水平明显上调(P<0.01)。相关性分析发现miR-708与JAK1表达呈负相关。结论 miR-708可能通过抑制其靶基因JAK1的表达在脑缺血再灌注损伤中发挥保护作用。 Objective To investigate the function and mechanism of expression changes of microRNA-708(miR-708) in rats with cerebral ischemia reperfusion injury. Methods Five male SD rats were randomly assigned to sham operation group (n = 10) and tMCAO group (middle cerebral artery infarct and reperfusion model, 10 rats survived after operation,n = 10). The miR-708 expression in brain tissues and blood and JAK1 mRNA expression in brain tissues of each group were detected by using real time polymerase chain reaction (qRT-PCR) method. The JAK1 protein level was detected by using Western blot method. Results Compared with those in sham operation group,miR-185 expressions were significantly lower (P〈0. 01) in brain tissues and blood and the expressions of JAK1 mRNA were significantly increased in brain tissues in the tMCAO group (P〈0. 01) )Correlation analysis found that the expression of miR-708 and JAK1 mRNA had negative correlation in the tMCAO rats. Conclusion MiR-708 may play a protective role in brain ischemia reperfusion injury by inhibiting the expression of the target gene JAK1.
