正交设计优选金刚藤总黄酮提取工艺研究 被引量:4

摘要 目的为分离纯化金刚藤总黄酮,筛选金刚藤药材最佳提取工艺。方法以提取干膏总黄酮含量、总黄酮转移率为指标,采用正交试验法,考察水提醇沉法提取加水量、提取时间、提取次数、醇沉浓度对工艺的影响,考察醇提法提取加醇量、乙醇浓度、提取时间、提取次数对工艺的影响;采用分光光度法测定干浸膏中总黄酮的含量,检测波长292 nm,落新妇苷对照品在4.04~24.24μg/mL浓度范围内呈良好的线性关系,平均回收率为98.52%,RSD为1.15%。结果最佳提取工艺为水提醇沉工艺,工艺参数为提取加水量10倍、提取次数2次、提取时间1.5 h和醇沉浓度70%。结论优选的提取工艺可行,可作为分离纯化金刚藤总黄酮的金刚藤药材提取工艺。 Objective To isolate and purify the total flavonoids from Smilax china L. and to optimize the extractionprocess. Methods With the content and transport rate of total flavonoids as indices, the effects of the amount ofwater, extraction time, extraction frequency and alcohol concentration on the process of water extraction and alcoholprecipitation were studied by orthogonal experiments. Meanwhile, the effects of the amount of alcohol, alcoholconcentration, extraction time and extraction frequency on the extraction process of alcohol extraction were alsostudied. The content of total flavonoids in dry extract was determined by spectrophotometry. The detection wavelengthwas 292 nm. There was a good linear relationship in the range of 4.04-24.24 μg/mL for astilbin control with theaverage recovery of 98.52 % and the RSD of 1.15 %. Results The optimum extraction process was water extractionand alcohol precipitation. The process parameters were as follows: the medical material was extracted for 2 times,once for 1.5 h after adding 10 times of water, and the last alcohol concentration was 70 %. Conclusion The optimumextraction process can be feasible and can be used for the separation and purification of total flavonoids from Smilaxchina L.
