结肠癌组织中转录因子Runx2表达量的变化及其与癌细胞生长和迁移的相关性 被引量:1

摘要 目的:探讨结肠癌组织中转录因子Runx2表达量的变化及其与癌细胞生长和迁移的相关性。方法:收集2014年12月~2017年5月在本院进行结肠癌根治术治疗的原发结肠癌患者90例作为结肠癌组,同期在本院进行肠镜检查的良性息肉患者80例作为息肉组。采用荧光定量PCR法检测不同病灶组织中转录因子Runx2,结肠癌相关增殖、侵袭基因表达量的差异,采用Pearson检验评估结肠癌组织中Runx2mRNA表达量与癌细胞生长迁移能力的相关关系。结果:结肠癌组织中Runx2mRNA的表达量高于息肉组;结肠癌组病灶组织中增殖基因MS4A12mRNA的表达量低于息肉组,GTPBP4、hTERT、CXCR7 mRNA的表达量高于息肉组;结肠癌组病灶组织中侵袭基因CCL21、LIMK1、PRDX1、STAT3、LIMK1、SLP-2mRNA的表达量高于息肉组。结论:结肠癌组织中转录因子Runx2表达量增加,与癌细胞增殖、侵袭活性增加直接相关。 Objective:To discuss the change of transcription factor Runx2 expression in colon cancer tissues and its correlation with cancer cell growth and migration.Methods:90 patients with primary colon cancer who received radical operation for colon cancer in the hospital between December 2014 and May 2017 were collected as colon cancer group,and 80 patients with benign polyps who received enteroscopy in the hospital during the same period were collected as polyp group.Fluorescence quantitative PCR method was used to detect the expression in transcription factor Runx2 as well as colon cancer-related proliferation and invasion genes in different lesion tissues,and Pearson test was used to evaluate the correlation between Runx2 mRNA expression and cancer cell growth migration in colon cancer tissues.Results:Runx2 mRNA expression in lesion tissue of colon cancer was higher than that of polyp group;proliferation gene MS4 A12 mRNA expression in lesion tissue of colon cancer group was lower than that of polyp group whereas GTPBP4,hTERT and CXCR7 mRNA expression were higher than those of polyp group;invasion genes CCL21,LIMK1,PRDX1,STAT3,LIMK1 and SLP-2 mRNA expression in lesion tissue of colon cancer group were higher than those of polyp group.Conclusions:Transcription factor Runx2 expression increases in colon cancer tissue and is directly correlated with the proliferation and invasion activity of cancer cells.
