摘要
为了获得重组CHO细胞培养的最佳条件,使其能连续高效表达HBsAg,以含不同量谷氨酰胺的DMEM溶液培养重组CHO细胞,测定了不同阶段氨基酸、氨、乳酸含量及HBsAg滴度。结果表明谷氨酰胺的分解产物氨与乳酸均能抑制细胞的生长和分泌HBsAg,减少细胞培养液中的谷氨酰用量可使HBsAg分泌量增加。
To obtain the best culture conditions of CHO cells and high efficiency secretion of HBsAg during the culture of recombinant CHO cells. DMEM solution that contain different glutamine volume was used in cell culture and detected the content of amino acid, amonia and lactic acid and HBsAg liter in different phase. The results showed that cells growth and secretion of HBsAg were limited by ammonia and lactic acid during the cell culture. Decreasing glutamine can increase the secretion of HBsAg in the cell culture.
