摘要
目的 观察雷公藤多甙 (TWP)、地塞米松 (DEX)对急性肺损伤 (ALI)的防治作用 ,并探讨其可能机理。方法 BLAB/C小鼠随机分成ALI组、TWP组、DEX组和生理盐水对照组。酶联免疫吸附法 (ELISA)测定脾细胞培养上清液中干扰素 (IFN) γ、白细胞介素 4 (IL 4 )蛋白含量。伊文思蓝荧光法测定肺血管蛋白渗出量。肺湿重 /干重比反映肺水量。HE染色观察肺病理变化 ,并对肺组织多形核粒细胞 (PMN)进行半定量计数。结果 (1)与对照组比较 ,ALI组脾细胞中IFN γ显著下降 (P <0 0 1)、IL 4明显升高 (P <0 0 1)。与ALI组比较 ,TWP组、DEX组脾细胞中IFN γ、IL 4均明显降低。 (2 )与ALI组比较 ,TWP组、DEX组肺组织伊文思蓝浸出量、肺湿重 /干重比和PMN计数均明显降低。结论 TWP、DEX对ALI均有防治作用 ,其中TWP以抑制抗炎症介质为主 ,对ALI以抗炎症反应为主时可能更为有益。相反 ,DEX抑制炎症介质为主 。
Objective To investigate the therapeutic effect of tripterygium wilfordii polyglyco sidium(TWP) and dexamethasone(DEX) on acute lung injury.Methods Mice were randomly divided into ALI group,saline control group,TWP group,DEX group.Splenocyte cytokine lnterferon γ,interleukin 4 in supernatant were quantified by enzyme linked immuno sorbent assay(ELISA).The pulmonary extravascular lung water was determined by lung wet to dry weight ratio.The quantity of protein immersed was measured by Evans blue fluorescence.The pathologic change of lung was detected by HE dying and Polymorphoneuclear(PMN) cells in lung were halfmeasured.Results As compared with control group,splenocyte cytokine IFN γ decreased( P< 0 01) whereas IL 4 increased( P< 0 01)in ALI group.Both IFN γ and IL 4 in TWP group and DEX group decreased significantly as compared to those of ALI group.Moreover,IL 4 levels in supernatant were significantly lower in TWP group than that in DEX group.On the contrary,IFN γ in supernatant were significantly lower in DEX group than that in TWP group.As compared with ALI group,the lung wet to dry weight ratio,PMN count in lung and quantity of lung Evans blue immersed were decreased markedly in DEX group and TWP group.Conclusions Both TWP and DEX can cure ALI.TWP has more inhibitory effect on anti inflammatory mediator,whereas DEX has more inhibitory effect on pro inflammatory mediator,both of which may be beneficial to restore balance of pro inflammatory and anti inflammatory response.
