摘要
通过静态吸附-解吸试验从3种吸附树脂中筛选出最适合分离谷胱甘肽(GSH)的树脂,将其与酵母渗透细胞耦合,通过补加前体物质,酶法合成GSH。结果表明,在试验的3种树脂中,D301吸附效果最佳。采用单因素和正交试验法,确定酶法合成GSH的优化工艺为:反应2.0 h后添加树脂2.0 g/10 mL,反应3.0 h时补加60%初始浓度的前体物质,最终得到GSH产量1 154 mg/L,与未添加树脂和前体合成法相比GSH产量提高77.5%。
The resin suitable for the separation of glutathione was screened from three kinds of adsorption resins by static adsorption-desorption experiments, the resin was coupled with yeast permeation cells, and enzymatic synthesize of glutathione was conducted by precursor addition. Results showed that the adsorption effect of resin D301 was the optimal among the three resins. The processing conditions were optimized by single factor and orthogonal experiments, in the enzymatic synthesis of glutathione experiments, adding resin 2.0 g/10 ml after reaction 2.0 h, and adding 60% initial concentration of precursor after reaction 3.0 h, finally the glutathione yield was 1 154 mg/L, which increased 77.5% compared with the method without resin and precursor addition.
