摘要
目的研究人肝癌衍生生长因子(HDGF)对Ewing肉瘤细胞生物学功能的影响。方法 HDGF-siRNAs沉默Ewing肉瘤细胞系RD-ES细胞中HDGF蛋白的表达,Western blotting检测蛋白表达的变化,克隆形成实验、细胞周期检测、Ed U细胞增殖实验和软琼脂克隆形成实验等方法用于分析内源性HDGF表达对Ewing肉瘤细胞增殖能力、细胞生长周期和体外肿瘤发生的影响。结果沉默内源性HDGF表达显著降低了Ewing肉瘤细胞的增殖能力,使细胞周期进程阻滞,并抑制了Ewing肉瘤细胞体外成瘤能力。结论 HDGF是Ewing肉瘤中重要的致瘤基因,在Ewing肉瘤细胞体外生长和肿瘤发生中发挥重要功能。
Objective To investigate the function role of hepatoma-derived growth factor (HDGF) in Ewing sarcoma cells. Methods HDGF protein level was down-regulated by specific siRNAs in RD-ES cells. The expression of HDGF protein was evalu- ated by western blotting analysis. Colony formation assay, cell cycle analysis, EdU cell proliferation assay, and soft agar colony for- mation assay were performed to examine the role of endogenous HDGF in proliferation, cell cycle progression and tumorigenesis in vitro of Ewing sarcoma cells. Results As compared with the negative control groups, depletion of endogenous HDGF strongly down- regulated cell proliferative ability, caused an cell cycle arrest, and inhibited the in vitro tumorigenic activity of RD-ES cells. Con- clusion Our findings strongly supported that HDGF acts as an oncogene in Ewing sarcoma and plays an important role in the growth and tumorigenesis of Ewing sarcoma cells in vitro.
