阿司匹林联合顺铂对食管癌Eca-109细胞增殖的抑制作用 被引量:2

摘要 目的探讨单药阿司匹林及其联合顺铂对食管癌Eca-109细胞增殖的抑制作用。方法将体外培养的Eca-109细胞随机分为空白对照组,阿司匹林不同浓度组(1.25、2.5、5、10、20 mmol/L),顺铂4.17μmol/L浓度组,5 mmol/L阿司匹林联合顺铂组,采用MTT法检测不同时间点(24、48、72 h)各组的细胞抑制率。结果同一时间点,不同浓度阿司匹林对Eca-109细胞的抑制率显著高于空白组(P<0.05),且呈浓度依赖性;在同一浓度下,细胞的抑制率随着作用时间的延长显著增加(P<0.05),即呈时间依赖性;同一时间点,阿司匹林联合顺铂对细胞的抑制率显著高于同浓度单药阿司匹林组(P<0.05)及单药顺铂组(P<0.05)。结论阿司匹林对食管癌Eca-109细胞的增殖有抑制作用,且呈浓度、时间依赖性,阿司匹林联合顺铂对Eca-109细胞具有协同抑制作用。 Objective To investigate the inhibitory effect of single drug aspirin and its combination with cisplatin on the proliferation of esophageal carcinoma Eca-109 cells. Methods The Eca-109 cells in vitro cultured were randomly divided into two groups:blank control group , aspirin groups (1.25 mmol/L,2.5 mmol/L,5 mmol/L, 10 mmol/L,20 mmol/L), cispla- tin (4.17 mmol/L) group and aspirin (5 mmol/L)combined with cisplatin group , Using MTI" method detect cells proliferation inhibition rate in each group at different time ( 24 hours,48 hours,72 hours). Results The inhibitory rate of each aspirin group on Eca-109 cells was significantly higher than that of the blank group at the same point of time (P 〈 0.05 ), and the pro- liferation inhibition rate of the Eca-109 cells was concentration-dependent. At the same degree of concentration, the proliferation inhibition rates were increasing with the increase of treatment time and showed time-dependent ( P 〈 0. 05 ). At the same time point, the inhibitory rate of aspirin combined with cisplatin was significantly higher than that of single drug aspirin group (P 〈 0. 05) and single cisplatin group (P 〈 0.05 ). Conclusion Aspirin inhibits the proliferation of esophageal cancer Eca-109 ceils in concentration and time dependence. Aspirin combined with cisplatin has a synergistic inhibitory effect on Eca-109 cells.
