摘要
目 的 观 察 并 分 析 抗 氧 化 剂 [维 生 素 C (vitaminC,VitC)、维 生 素 E (vitaminE, VitE)]对支气管哮 喘 (简 称 哮 喘 ) 大 鼠 气 道 重 塑 的 作 用, 对 血 浆 转 化 生 长 因 子 β1 (transforming growthfactor-β1,TGF-β1)水平表达变化的影响,以及分析 TGF-β1 与气道重塑的相关性。方法 将50只大鼠随机分组,哮喘组用卵清蛋白经典方法诱发哮喘,连续诱发6周后处死大鼠,取肺组织给予 HE染色和 Masson染色,观察各 组 大 鼠 气 道 重 塑 及 纤 维 化 的 情 况,测 血 TGF-β1 浓 度,进 行 统 计 分 析。结果 抗氧化剂 VitC、VitE可使哮喘大鼠的支气管管壁厚度、平滑肌厚度、胶原纤维组织面积减 小,减少血浆 TGF-β1 的表达;联合组的气道重塑较 VitC 组轻,但与 VitE 组差异无 统 计 学 意 义,联 合应用时抗氧化作用强于单用,但与 VitE 组差别不大;VitC 联合 VitE 对气道重塑的缓解作用最强,但不能认为其可完全缓解气道重塑;Perman相关性分析得出 TGF-β1 表达水平与气道重塑呈正相 关。 结论 抗氧化剂 VitC、VitE可缓解气道重塑,对哮喘的治疗 及 哮 喘 气 道 重 塑 的 预 防 有 一 定 作 用;血 TGF-β1 表达水平可在一定程度上反映气道重塑,可作为气道重塑的重要检测因子;VitC 与 VitE 之间 没有相互促进作用。
Objective To observe and analyze the antioxidants (vitamin C, vitamin E) the role of airway remodeling of asthma rats, the expression of transforming growth factor-β1 (TGF-β1) level changes, as well as the analysis of TGF -β1 and the correlation of airway remodeling. Methods Fifty rats were randomly groups, asthma group of asthma induced by injection of ovalbumin classical method, after six weeks of continuous induction, rats were executed, take the lung tissue to HE and Masson staining of eolor, observe each group of rats with airway remodeling and fibrosis, detect concentration of TGF-β1 and statistical analysis. Results Antioxidants vitamin C and vitamin E make decrease the thickness of the bronchial wall, the smooth muscle and collagen fibre area of asthma rats, reduce the expression of TGF- β1, Joint group of airway remodeling is lighter vitamin C group, but there was no statistically significant difference with vitamin E group, The joint appliation antioxidant effect than when alone, but there was so little difference between vitamin E group, vitamin C joint vitamin E strongest relief of airway remodeling, but cannot think it can eompletely relieve the airway remodeling. Perman correlation analysis TGF-β1 expression level was positively related with airway remodeling. Conclusions Antioxidants vitamin C and vitavnin E can alleviate airway remodeling, treatment of asthma and the prevention of airway remodeling of asthma have a certain role. The expression of TGF-β1 level reflects the airway remodeling in a certain extent, can be used as the airway remodeling of important detection factor. There is no mutual promoting effect between vitamin C and vitamin E.
