营心丹对大鼠急性心肌缺血预防性保护作用研究

Preventive protection of Yingxindan on myocardial ischemia

目的观察营心丹对垂体后叶素(pituitrin,pit)诱导的急性心肌缺血预防性保护作用。方法60只大鼠随机分为硝苯地平12.6 mg·kg^(-1)组、营心丹高、中、低剂量组(25.2、12.6、6.3 mg·kg^(-1))、生理盐水组和模型组(给予相同体积的纯水),灌胃给药,连续给药7 d,最后1次给药1 h后,舌下静脉注射pit 1 U·kg^(-1)造成急性心肌缺血模型。记录注射后10 min内不同时间点的肢体Ⅱ导联心电图ST段、心肌缺血阳性率及心律失常的变化,考察营心丹对血清中肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、糖原磷酸化同工酶BB(GPBB)、心肌肌钙蛋白I(c Tn I)、心肌肌钙蛋白T(c Tn T)的影响。结果营心丹对垂体后叶素所致急性心肌缺血大鼠心电图ST段变化有明显的改善作用,降低心肌缺血阳性率及心律失常发生率;明显降低血清中心肌损伤标志物CK、CK-MB、GPBB、c Tn I、c Tn T的含量,营心丹剂量越大,血清中心肌损伤标志物含量越小。结论营心丹对垂体后叶素致大鼠急性心肌缺血具有明显的预防性保护作用。

Aim To observe the preventive protection of Yingxindan on acute myocardial ischemia induced by sublingual injection of pituitrin(pit)in rats.Methods Sixty rats were randomly divided into saline group,model group,nifedipine 12.6 mg·kg^(-1)group,and Yingxindan 25.2,12.6,6.3 mg·kg^(-1)groups.The saline group and model group were given the same volume of water.After administration for 7 d,an acute myocardial ischemia model was induced by sublingual intravenous injection of pit 1 U·kg^(-1)in the rats 1 h after last administration.The changes of the II electrocardiogram(ECG)ST segment at different time points,as well as the positive rate of myocardial ischemia and arrhythmia were observed.Meanwhile,the levels of serum creatine kinase(CK),creatine kinase isoenzyme(CK-MB),glycogenphosphorylase isoenzyme BB(GPBB),cardiac troponin I(c Tn I)and cardiac troponin T(c Tn T)were observed.ResultsYingxindan could suppress the changes of ST segment of electrocardiogram in rats with acute myocardial ischemia induced by pit and reduce the positive rate of myocardial ischemia and arrhythmia.The contents of myocardial injury markers CK,CK-MB,GPBB,c Tn I and c Tn T in serum significantly decreased,and the corresponding relationship existed.The greater the dose of Yingxindan,the smaller the content of myocardial injuried markers in serum.Conclusion Yingxindan has significant preventive protection effect on acute myocardial ischemia by pit.
