摘要
目的 :观察大鼠全脑缺血再灌注后前脑NOS阳性神经元分布和FOS蛋白表达时程变化 ,探讨其相互间的变化关系及其在脑缺血损伤机制中的可能作用。方法 :采用四血管闭塞模型、运用NADPHd组化反应、FOS免疫组化反应及两者的双重反应技术 ,对全脑缺血 30min再灌注 2h、4h、8h、1 2h、2 4h、4 8h进行研究。结果 :缺血组前脑NOS阳性神经元数目和染色强度增加 ,FOS蛋白表达随再灌注时程而变化 ,并可见有约 1 0~ 1 5 %NOS/FOS双染细胞。结论 :全脑缺血再灌注损伤过程中 ,前脑NOS阳性神经元的分布与FOS蛋白表达密切相关。
Objective:To observe the changes of NOS distribution and time course of FOS protein expression in different regions of rat forebrain following cerebral ischemia-reperfusion. Methods: NADPH-d histochemical and FOS immunohistochemiacal method were used to explore the reperfusion of 2 h,4 h,8 h,12 h,24 h,48 h after ischemia 30 min in rats model of 4 vessel occlusion. Results: The density of NOS postive neurous and its containing reaction increased in the ischemia groups. FOS protein expression was found with the changes of the time after ischemia reperfusion. Approximately 10%~15% of the NOS/FOS double labelled cells showed in several regions. Conclusion: The results indicate that the distribustion of Nos positive necrons and Fos protein expression are closely related to the reperfusion injury after ischemia.
