双环醇对肺间质纤维化小鼠TGF-β_1表达的影响 被引量:1

摘要 目的采用盐酸博来霉素建立小鼠肺间质纤维化模型,观察双环醇对小鼠肺间质中转化生长因子-β1(TGF-β1)表达的影响。方法将54只小鼠随机分为正常对照组、模型组、双环醇干预组,各18只。模型组和双环醇干预组小鼠经乙醚麻醉后经气管滴入盐酸博来霉素(5μg/g),建立肺间质纤维化动物模型。双环醇干预组于模型建立1天后予双环醇10mg/kg灌胃,正常对照组与模型组予等量生理盐水灌胃,于干预后第7、14、28天3个时间点各组随机处死6只小鼠,取肺组织进行病理学观察并按照Szapiel方法评价肺泡炎和肺纤维化程度,免疫组化染色观察TGF-β1蛋白的表达。结果模型组干预后第7、14、28天肺间质损伤评分分别为3.37±0.80、5.03±0.75、6.12±1.09,而双环醇干预组分别为2.22±0.73、3.86±0.79、5.24±1.05,双环醇干预组明显低于模型组(P<0.05)。模型组干预后第7、14、28天肺组织TGF-β1的相对表达分别为9.67±1.92、12.53±1.43、14.34±1.43,而双环醇干预组分别为5.02±1.27、8.45±1.19、10.81±1.73;双环醇干预组明显低于模型组(P<0.05)。结论双环醇对小鼠肺间质纤维化具有显著的改善作用,可减轻小鼠肺间质纤维化程度,其机制可能与双环醇下调TGF-β1的表达有关。 Objective The purpose of this study is to explore the effects of bicyclol on the pulmonary fibrosis(PF) in rats with TGF-β1, and to observe the effects of bicyclol on the expression of pulmonary fibrosis(PF), so as to provide a new train of thought for the treatment of pulmonary fibrosis. Method The rats were treated with diethyl ether and then administered to BLMA5, 5 mg/g through the weasand so as to the animal model of pulmonary fibrosis were established. 54 rats were randomly divided into model group(n=6) and bicyclol intervention group(n=6), normal control groups(n=6) also were established. According to the above methods model group and bicyclol intervention group copy the animal model. Bicyclol intervention group was intragastric administration(ia) at bicyclol 10 mg/kg once a day. In each group, six rats were chosen randomly to be killed at the 7 d, 14 d and 21 d after operation for histological examination of lung tissue. After that evaluate alveolitis and pulmonary fibrosis degree with Szapiel method. The expression of TGF-β1 was detected by immunohistochemistry and in-situ hybridization. Result The model group after the third day 7,14,28 interstitial lung injury score was 3.37+0.80, 5.03+0.75, 6.12+1.09, and bicyclol intervention group were2.22 + 0.73, 3.86 +0.79, 5.24+1.05, bicyclol intervention group was significantly lower than the model group(P〈0.05). The model group dry day 7,14,28 relative expression of prognosis of lung group fabric TGF-β1 were 9.67+1.92, 12.53+1.43, 14.34+1.43, and bicyclol intervention group were 5.02+1.27, 8.45+1.19, 10.81+1.73; bicyclol intervention group was significantly lower than the model group(P 0.05). Conclusion Bicyclol has the predominant improve effect to the pulmonary fibrosis in bleomycin hydrochloride rats, which maybe thought the promote by down-regulating the expression of TGF-β1.
