摘要
为探讨高原低氧环境下大通犊牦牛线粒体琥珀酸脱氢酶(SDH)在不同组织中的表达模式以及SDH活性与其亚基mRNA表达量之间的关系,采用酶联免疫分析法(ELISA)对犊牦牛心肌、骨骼肌、大脑、肝脏组织中线粒体SDH活性进行测定;实时荧光定量PCR(Real-Time PCR)检测组织中Sdha、Sdhb、Sdhc和Sdhd亚基mRNA表达量。结果表明:大通犊牦牛大脑、心肌、骨骼肌组织中线粒体SDH活性显著高于乐都犊牦牛;大脑组织中线粒体Sdh4个亚基mRNA表达量极显著高于乐都犊牦牛。结论:含野血大通犊牦牛组织线粒体SDH有氧代谢功能强于乐都犊牦牛;大脑组织线粒体Sdh4个亚基转录水平较高;大通犊牦牛大脑组织线粒体SDH活性与Sdh4个亚基mRNA表达量呈正相关。
Expression patterns of mitochondrial succinate dehydrogenase(SDH) in different tissues of Datong infant yak, as well as thecorrelation among SDH and mRNA expression quantities ofSdha,Sdhb,Sdhc andSdhd, were investigated under the plateau hypoxia envi-ronment. The cardiac muscle, skeletal muscle, liver and brain samples were collected from infant yak, and the activity of mitochondrial SDHwere analyzed by enzyme-linked immunoassay(ELISA), mRNA expression quantities ofSdha,Sdhb,Sdhc andSdhd were determined byqRT-PCR. The results showed that mitochondrial SDH activities of the cardiac muscle, skeletal muscle and brain in Datong infant yak weresignificantly higher than those in Ledou infant yak, and mRNA expression quantities ofSdha,Sdhb,Sdhc andSdhd of cerebral mitochondriain Datong infant yak were remarkably higher in relative to those in Ledou infant yak. In conclusions, aerobic metabolism of mitochondrialSDH in Datong infant yak with wild yak blood was more active compared to that in Ledou yak, and the mRNA expression levels ofSdha,Sdhb,Sdhc andSdhd subunits were up-regulated in the brain of Datong infant yak. Moreover, the level of SDH in cerebral mitochondria ofDatong infant yak was positively correlated with the abundance ofSdha,Sdhb,Sdhc andSdhd mRNA expressions.
