摘要
为探究Pur诱导hPSCs向基底前脑抑制性中间神经元分化的最佳浓度。以前期建立的基底前脑γ-氨基丁酸(gammaaminobutyric acid,GABA)能中间神经元分化方法为基础,在分化过程中加入不同浓度的Pur,应用免疫荧光、PCR等技术探究不同浓度的Pur对GABA能中间神经元的分化作用。2.0μmol/L的Pur分化可获得纯度较高的GABA能中间神经元,而1.5μmol/L的Pur分化可获得数量较多的GABA能中间神经元。高浓度的Pur能够有效诱导hPSCs向基底前脑分化,并高效地分化为前脑GABA能中间神经元。
Based on the protocol,the forebrain GABAergic interneurons from hPSCs were differentiated,and the GABAergic interneurons with different dosages of Pur were added.The results were analyzed by immunofluorescence and PCR for the effect of patterning hPSCs to GABAergic interneurons with different dosages of Pur.The results show that after applying 2.0μmol/L Pur,higher purity of GABAergic interneurons can be obtained.After applying 1.5μmol/L Pur,more GABAergic interneurons are attained.Our study indicated that higher concentration of Pur can effectively induce hPSCs to basal forebrain and differentiate into forebrain GABAergic interneurons.
