摘要
目的探讨二乙酰二去水卫矛醇(DADAG)对人鼻咽癌细胞HNE-1、人肺癌细胞A549、人肝癌细胞Hep G2、SMCC-7721及胃癌细胞MGC-803的体外抑制作用。方法采用不同浓度DADAG干预处于对数生长期的HNE-1、A549、Hep G2、SMCC-7721、MGC-803细胞,其中A549、Hep G2、SMCC-7721细胞的药物干预浓度包括0μg/ml(空白组)、2.88μg/ml、5.75μg/ml、11.50μg/ml、23.00μg/ml、46.00μg/ml,MGC-803、HNE-1细胞的干预浓度包括0μg/ml(空白组)、11.50μg/ml、23.00μg/ml、46.00μg/ml、69.00μg/ml、92.00μg/ml。显微镜下观察经DADAG作用后的细胞形态,采用噻唑蓝比色法检测5种肿瘤细胞的增殖情况,计算5种细胞的增殖抑制率及DADAG对5种细胞的半抑制浓度(IC50)值。结果经DADAG作用后,5种细胞的形态发生不同程度改变。除11.50μg/ml DADAG作用下的MGC-803细胞外,不同浓度DADAG作用下各细胞的A值均低于空白组(P<0.05)。随着给药浓度的增加,5种肿瘤细胞抑制率也逐渐增加。各给药浓度下,5种细胞间的A值比较,差异均有统计学意义(P<0.05)。DADAG对SMCC-7721、MGC-803、HNE-1、A549、Hep G2的48 h-IC50值分别为145.39μg/ml、141.29μg/ml、49.36μg/ml、47.75μg/ml、28.52μg/ml。结论 DADAG在体外对上述5种癌细胞均有抑制生长作用;肝癌细胞SMMC-7721及胃癌细胞MGC-803对DADAG敏感性较其他细胞差。
Objective To investigate the inhibitory effect of diacetyldianhydrogalactitol( DADAG) on human nasopharyngeal carcinoma cell HNE-1,human lung cancer cell A549,human liver cancer cell HepG2 and SMCC-7721,and human gastric cancer cell MGC-803 in vitro. Methods Intervention using DADAG of different concentrations was conducted in HNE-1,A549,HepG2,SMCC-7721 and MGC-803 cells in the logarithmic phase. The concentrations of DADAG for A549,HepG2 and SMCC-7721 cells included 0 μg/ml( blank group),2. 88 μg/ml,5. 75 μg/ml,11. 50 μg/ml,23. 00 μg/ml and 46. 00 μg/ml,and the concentrations for MGC-803 and HNE-1 cells included 0 μg/ml( blank group),11. 50 μg/ml,23. 00 μg/ml,46. 00 μg/ml,69. 00 μg/ml and 92. 00 μg/ml. The cellular morphology after intervention of DADAG was observed under microscope. The proliferation of 5 types of tumor cells was detected using methylthiazolyldiphenyltetrazolium bromide assay. The inhibitory rates of 5 types of tumor cells and half maximal inhibitory concentrations( IC50) of DADAG for 5 types of tumor cells were calculated. Results Different changes in morphology were observed in 5 types of tumor cells after intervention of DADAG.Except MGC-803 cells under intervention of 11. 50 μg/ml DADAG,A value of each type of the cells was lower than that of the blank group under the intervention of DADAG with different concentrations( P〈0. 05). The inhibitory rates of 5 types of tumor cells increased with the increase of the drug concentrations. There was significant difference in A value among 5 types of tumor cells( P〈0. 05). The 48 h-IC50 of DADAG for SMCC-7721,MGC-803,HNE-1,A549 and HepG2 cells were 145. 39 μg/ml,141. 29 μg/ml,49. 36 μg/ml,47. 75 μg/ml and 28. 52μg/ml,respectively. Conclusion DADAG obtains a growth-inhibiting effect on the 5 types of tumor cells above in vitro. Liver cancer cell SMCC-7721 and gastric tumor cell MGC-803 are less sensitive to DADAG compared to the other cells.
