Determination of betaine in dried wolfberry by ion chromatography with pulsed amperometric detection

摘要 建立了一种离子色谱结合脉冲安培检测器测定干制枸杞中甜菜碱的方法。以水为提取溶剂对干制枸杞进行萃取,经过滤直接作为待测样品分析,以阴离子交换树脂为填料的色谱柱,以甲基磺酸作为流动相对干制枸杞中的甜菜碱进行了测定。10 min内完成了高效分离。方法线性关系良好,回收率在83.2%~86.7%,精密度2.45%,检出限0.0366 mg/L。该方法只使用价格低廉的纯水作为提取溶剂,避免了使用有毒有害的有机溶剂,可用于功能性食品中甜菜碱的快速、批量检测。 A chromatographic method by ion chromatography with pulsed amperometric detection was developed for the determination of the betaine in dried wolfberry. The betaine in dried wolfberry sample was conveniently extracted with water as extraction solvent and directly used as the sample to be analyzed after filtrating. The wolfberry can be separated well in 10 min by high performance anion exchange column(HPAEC) with methanesulfonic acid(MAS) as the mobile phase. The detection limit of betaine(10 m L injection, S/N=3) using pulsed amperometric detection(PAD) was 0.0366 mg/L. Moreover, the good relative standard deviations(2.45%) and satisfying recoveries(83.2%~86.7%) were obtained. This method only utilized water as an extraction solvent with low cost, and avoided the use of toxic and hazardous organic solvents, which provided a quite suitable platform for batch and fast testing of betaine in plant and functional foods.
