摘要
目的探讨雷帕霉素对人口腔上皮癌耐药细胞株KB/VCR的多药耐药逆转作用及其逆转机制。方法采用四甲基偶氮唑盐(MTT)方法检测KB/VCR细胞中雷帕霉素对VCR的逆转倍数,流式细胞仪检测Rh123在KB和KB/VCR细胞内的蓄积,Western-blot检测雷帕霉素对P糖蛋白(P-gp)表达的影响。结果雷帕霉素显著逆转KB/VCR的耐药性。雷帕霉素能够明显增强Rh123在细胞内的蓄积,同时能够降低P-gp的表达。结论雷帕霉素能够逆转KB/VCR细胞对VCR的耐药性,可能与其抑制P-gp功能和蛋白质表达相关。
Objective To explore the reversal effect and potential mechanism of rapamycin on multidrug resistance of KB/VCR cells. Methods MTT assay was used to investigate reversal index of rapamycin to vincristine. P-gp activity was evaluated by measuring Rh123 accumulation by flow cytometry. Western blot was used to detect the protein expression of P-gp. Results Rapamycin obviously reversed the multidrug resistant phenotype of KB/VCR cells. Further study showed that rapamycin significantly increased the accumulation of Rh123 in KB/VCR cells. Moreover,rapamycin down-regulated the expression of P-gp. Conclusion Rapamycin significantly reverse P-gp-mediated MDR in KB/VCR cells. The potential mechanism may be via inhibiting the activity and expression of P-gp.
