摘要
目的研究葛根素对人胆管癌QBC939细胞生长抑制的影响。方法首先体外培养人胆管癌QBC939细胞株,应用不同浓度葛根素处理QBC939细胞株,应用CCK8法检测不同浓度葛根素对胆管癌QBC939细胞的生长抑制作用;应用RT-PCR法检测葛根素作用于胆管癌QBC939细胞后细胞中血管内皮生长因子-C(VEGF-C)、环氧合酶-2(COX-2)基因表达变化。结果 (1)CCK8法检测结果显示,葛根素对胆管癌QBC939细胞的生长具有明显抑制作用,且随着浓度的增加,其对胆管癌QBC939细胞的抑制作用越明显;(2)RT-PCR法显示葛根素明显抑制胆管癌QBC939细胞中VEGF-C,COX-2基因表达。结论葛根素对人胆管癌QBC939细胞的生长有显著抑制作用。
Objective To observe the inhibitory effect on growth of human cholangiocarcinoma QBC939 cell treated by puerarin.Methods In vitro cultured human cholangiocarcinoma QBC939 cells were given different concentrations of puerarin. The relative ratio of QBC939 cells proliferation restraint was measured by the CCK8 kit. The expression of vascular endothelial growth factor-c( VEGF-C) and cyclooxygenase-2( COX-2) in cholangiocarcinom was determined by RT-PCR technique. Results(1)The CCK8 method showed that puerarin had obviously inhibitory effect on cholangiocarcinoma QBC939 cell,and with the increase of concentration,the inhibitory effect was more obvious.(2)The RT-PCR method showed that puerarin significantly inhibited the expression of VEGF-C and COX-2 genes in cholangiocarcinoma QBC939 cells. Conclusion Puerarin has significant inhibitory effecton human cholangiocarcinoma QBC939 cell.
