摘要
目的探讨重症抑郁症预后与circRNA表达水平、社会心理因素的关系。方法采用基因芯片技术筛查抑郁症患者与健康者间差异表达的circRNA,运用q PCR检测30例抑郁症患者外周血circRNA表达水平;并用汉密尔顿抑郁量表(HAMD)、中国社会支持量表(CSSS)、特质应对方式问卷(TCSQ)对患者进行评估。结果 circRNA_104600的△CT值与焦虑/躯体化、绝望感减分率呈负相关(P<0.05)。CSSS中客观支持因子分与HAMD绝望感减分率呈负相关(P<0.05);支持利用度因子分与HAMD总分、认知障碍、睡眠障碍的减分率呈正相关(P<0.05);TCSQ中消极应对因子分与HAMD总减分率呈负相关(P<0.05);积极应对因子分与焦虑/躯体化、体重减分率呈正相关(P<0.05)。circRNA_103964、CSSS中支持利用度对HAMD总减分率有预测作用(P<0.05),可解释因变量变异的64.1%;circRNA_100018、circRNA_104121、circRNA_104600、circRNA_103964对HAMD焦虑/躯体化减分率有预测作用(P<0.01),可解释因变量变异的88.0%;circRNA_103964对HAMD认知障碍减分率有预测作用(P<0.05),可解释因变量变异的57.0%;circRNA_103964、CSSS中支持利用度、TCSQ中积极应对对HAMD绝望感有预测作用(P<0.05),可解释因变量变异的74.0%。结论抑郁症预后与circRNA_104600、circRNA_103964表达水平、客观支持、支持利用度及积极应对方式有密切的关系。
Objective To explore relationship between prognosis of major depressive disorder ( MDD) and circRNA expression level, social psychological factors. Methods Different expressions of circRNA between patients with MDD and healthy persons were screened through Agilent Human circRNA. 30 patients with MDD were detected for peripheral blood circRNA expression level through qPCR. They were assessed with Hamilton Depression Scale (HAMD) , Chinese Social Support Scale( CSSS) and Trait Coping Style Questionnaire(TCSQ) . Results A C T value of circRNA_104600 was negatively correlated with reducing rates of anxiety/somatization and hopelessness( P 〈 0. 05 ) . Factor score of objective support of CSSS had negative correlation with reducing rate of hopelessness of HAMD (P 〈 0 . 05 ) ; meanwhile factor scores of support utility of CSSS had positive correlation with reducing rates of HAMD, cognitive disorder, sleep disorder(P 〈0.05). Negative coping style of TCSQ was negatively correlated with reducing rate of HAMD(P 〈0. 05) ; and positive coping style of TCSQ was positively correlated with reducing rates of anxiety/somatization and body weight ( P 〈0. 05 ) . Regressive analysis suggested that A C T value of circRNA_l03964, support utility of CSSS had significant predictive effect on reducing rate of HAMD ( P 〈 0. 05 ) and could account for 64. 1% variance. ACT values of circRNA_100018, circRNA_104121, circRNA_104600, circRNA_103964 had predictive effect on reducing rate of anxiety/somatization(P 〈0. 01) and could explain 88.0% variance. ACT value of circRNA _103964 had predictive effect on reducing rate of cognitive disorder(P 〈0.05) and could explain 57. 0% variance. ACT value of circRNA_l 03964, support utility of CSSS and positive coping style of TCSQ were significant predictors of hopelessness of HAMD (P 〈 0. 05 ) and could account for 74. 0% variance. Conclusion Prognosis of MDD is closely related with expression levels of circRNA_104600,circRNA_103964, objective support, support utility and positive coping style.
