一株鸭源H_9亚型禽流感病毒的分离鉴定及HA基因序列分析

摘要 本研究从发生产蛋下降疫情的山东某种鸭场采集病料,分离得到一株病毒。经血凝(HA)试验、血凝抑制(HI)试验、测序分析,鉴定该毒株为H_9亚型禽流感病毒。对分离株HA基因进行测序及序列分析,结果显示该分离株HA基因的裂解位点为333PARSSR↓GLF339,符合低致病性AIV的特征;与参考毒株的HA基因组序列进行比较发现,核苷酸的同源性为93.7%~99.3%,推导的氨基酸的同源性为96.4%~98.6%,与Shandong/YT5/2010(H_9N_2)株同源性最高。 Samples were collected from a breeding duck farm with egg-laying epidemic in shandong province,and one virus strain as isolated.The virus was identified as H9 subtype AIV by HA,HI test and sequencing analysis of HA gene.The sequencing analysis showed that the isolate had a HA cleavage site of 333 PARSSR ↓ GLF339,which was a characteristic structure of low pathogenic AIV. The nueleotide homology and amino acid homology between isolated strain and the reference strains were 93.7% 99.3% and 96.4% 98.6% respectively.The isolated strain was highest homology with Shandong/YT5/2010 H9N2 subtype.
