摘要
采用高效液相二极管阵列检测法测定延胡索提取物中延胡索乙素含量。采用ZORBAX XDB-C18色谱柱(4.60 mm×250 mm,5μm),以0.1%磷酸水溶液和甲醇为流动相,流速1.0 mL/min,柱温30℃,检测器温度30℃,检测波长280 nm。精密度和稳定性良好,延胡索乙素质量浓度为5~700 mg·L^(-1),标准物质峰面积与质量浓度线性关系良好(r>0.999 5);加标回收率为98%~102%,相对标准偏差为0.94%~1.31%。
A method for determination of tetrahydropalmatine in yanhusuo extract by HPLC was developed. An XDB-CI8 column (4.60 mm-250 mm, 5 μm) was used with methanol-0.1% phosphoric acid aqueous solution as binary mobile phase and the flow rate was 1,0 mL/min. The wavelengths for detection were set at 280 nm and the temperatures of the column and the detector were both 30℃. The precision and stability were fine. Linear correlation (r〉0.999 5) was observed between the peak area of standard substance and the mass concentration of tetrahydropalmatine in the concentration range of 5-700 mg·L-1. The recoverties of tetrahydropalmatine were 95%-102% with RSD values between 0.94% to 1.31%,
