摘要
为探索稻曲球不同储存时期的稻曲病菌分离最佳方法,采用新鲜稻曲球和储存7个月的稻曲球为材料,分别采取厚垣孢子悬液法和组织块分离法进行稻曲病菌的分离。结果表明:新鲜稻曲球厚垣孢子悬液法分离成功率最高达95.1%,且黄色厚垣孢子分离成功率高于墨绿色的老熟厚垣孢子;组织块分离法分离成功率最高为9.7%。储存7个月的稻曲球采用厚垣孢子悬液法分离成功率为0,改进后的组织块分离法分离成功率最高达55.1%,升汞消毒时间40 s为最佳。
To optimize isolation methods for Ustilaginoidea virens in different storage periods, patho- gens of Ustilaginoidea virens were isolated by chlamycladosporal suspension and body tissue. The re- sults showed that isolation rate of single spore from fresh yellow chiamydospore reached 95.1% with chlamycladosporal suspension, and it was higher than that of atroviren. But isolation rate of single spore from body tissue only reached 9.7%. The method of chlamycladosporal suspension for balls which had been stored for 7 months was defeated. 55.1% was reached successfully with the modified methods and the best disinfection time with mercury chloride was 40 s.
