姜三七对子宫肌瘤大鼠子宫组织细胞凋亡及Bcl-2、Bax蛋白表达的影响 被引量:9

Effects of Ginger Notoginseng on the Apoptosis and Expressions of Bcl-2 and Bax in the Uterine Tissue of Rat with Hysteromyoma

摘要 目的:探究姜三七对子宫肌瘤大鼠子宫组织细胞凋亡及Bcl-2、Bax蛋白表达的影响。方法:选取SPF级雌性SD大鼠60只,随机分为对照组、模型组、姜三七组,每组20只。模型组和姜三七组给予黄体酮注射液和苯甲酸雌二醇注射液腹腔注射,16周后建立大鼠子宫肌瘤模型,对照组给予等量生理盐水腹腔注射。模型建立后,姜三七组予0.1 g/ml的姜三七混悬液10 ml/kg灌胃,正常对照组和模型组给予等量生理盐水,每日一次,4周为一个疗程,治疗3个疗程。治疗结束后,对比各组大鼠子宫湿重、子宫系数、子宫组织ER(雌激素受体)、PR(孕激素受体)、细胞凋亡率以及Bcl-2、Bax蛋白的表达情况。结果:(1)与对照组比较,模型组大鼠的子宫湿重、子宫系数明显升高,而姜三七组大鼠子宫湿重、子宫系数明显低于模型组,差异均有统计学意义(P<0.05);(2)与对照组(30%)比较,模型组大鼠子宫组织ER的阳性表达率(90%)明显较高,姜三七组大鼠子宫组织ER的阳性表达率(75%)明显低于模型组,差异均有统计学意义(P<0.05);(3)模型组大鼠子宫组织PR的阳性表达率(80%)明显高于对照组(30%),而姜三七组大鼠子宫组织ER的阳性表达率(35%)明显低于模型组,差异有统计学意义(P<0.05);(4)模型组大鼠子宫组织细胞凋亡率低于对照组、Bax阳性细胞表达明显低于对照组,Bcl-2的阳性表达明显高于对照组,姜三七组大鼠子宫组织Bax的阳性表达与模型组比较明显增强,Bcl-2的阳性表达及细胞凋亡率明显降低,差异有统计学意义(P<0.05)。结论:姜三七能够降低子宫肌瘤大鼠子宫组织细胞凋亡、ER、PR及Bcl-2的表达,增强Bax蛋白表达。 Objective: To explore the effects of ginger notoginseng on the apoptosis and expressions of Bcl-2 and Bax protein of uterine tissue in rats with hysteromyoma. Methods: 60 female SPF SD rats were selected and randomly divided into the control group, model group and ginger notoginseng group, 20 rats in each group. Model group and ginger notoginseng group were given progesterone injection and intraperitoneal injection of estradiol benzoateinjection for 16 weeks to induce uterine leiomyoma in rat. The control group was given normal saline intraperitoneal injection. After the establishment of model, rats in the ginger notoginseng group and the other two groups were given I0 ml/kg of ginger notoginseng suspension (0.1 g/ml) and normal saline, respectively by gavage once a day, 4 weeks was considered as a treatment course, 3 courses of treatment in total. After the treatment, the uterine wet weight, uterine coeffi- cient, uterine tissue of ER (estrogen receptor) and PR(progesterone receptor), Bcl-2 protein and Bax protein expressions were compared between different groups. Results: ① The wet weight of uterus and uterus coefficient in rats of model group were significantly higher than those of the control group and in ginger notoginseng group (P〈0.05). ②The positive expression rate of ER in model group (90%) was significantly higher than that of the control group (30%) and the ginger notoginseng group (75%)(P〈0.05). ③The positive expression rate of PR in model group (80%) was significantly higher than that in the control group (30%) and ginger notoginseng group (35%) (P〈0.05). ④The positive expression of Bax and the apoptotic rate were significantly decreased but the positive expression of Bcl-2 was significantly increased in rats of model group than that in control group (P〈0.05). Compared with the model group, the positive expression of Bax protein in rats of ginger notoginseng group was obviously increased, but the expression of Bcl-2 and apoptotic rate were significantly decreased (P〈0.05). Conclusion: Ginger notoginseng could decrease the expressions of ER, PR and Bcl-2 protein in rat with hysteromyoma, but increase the expression of Bax.
