补肾活血通腑方对慢性脑供血不足模型大鼠SIRT1信号通路下蛋白表达的影响 被引量:3

摘要 目的:探讨补肾活血通腑方对慢性脑供血不足(CCCI)大鼠脑组织的保护作用。方法:雄性健康Wistar大鼠50只,随机分为五组:假手术组、模型组、尼莫地平组、补肾活血通腑方低剂量组及高剂量组,采用2-VO永久结扎剪断方法制备CCCI大鼠模型。大鼠连续给药灌胃8周之后,断头取海马,运用Western blot法进行蛋白检测。结果:与假手术组相比,模型组和其他治疗组在额叶PGC-1α及FOXO3α蛋白表达明显下降(P<0.05)。与模型组相比,补肾活血通腑方低剂量组海马的PGC-1α蛋白表达明显增加(P<0.05);与尼莫地平组相比,补肾活血通腑方低剂量组FOXO3α、PGC-1α蛋白表达明显增加(P<0.05)。结论:CCCI大鼠的脑缺血可以抑制FOXO3α、PGC-1α蛋白表达;补肾活血通腑方低剂量对FOXO3α、PGC-1α蛋白的表达有改善作用,增加脑供血,对CCCI大鼠脑组织有保护作用。 Objective:To study the protective effect of Bushen Huoxue Tongfu formula(BHTF) on brain tissue of rats with chronic cerebral circulatory insufficiency (CCCI). Methods:Fifty healthy male Wistar rats were randomly divided into five groups:sham operation group,model group,nimodipine group,BHTF of low dose group,BHTF of high dose group. The CCCI rat model was established using 2-VO permanent ligation. After 8 weeks of intragastric administration,the rats were sacrificed through decapitation,and the hippocampus was harvested. The protein was detected by Western blot. Results:Compared with the sham group,the expression of PGC-1α and FOXO3α in the frontal lobe of the model group and other treatment groups were decreased significantly(P〈0.05). Compared with the model group,the expression of PGC-1α protein in the hippocampus of the BHTF of low dose group was significantly higher than that in the model group (P〈0.05). Compared with the nimodipine group,FOXO3α,PGC-1α protein expressions were significantly increased (P〈0.05). Conclusion:Cerebral ischemia in CCCI rats can inhibit the expressions of FOXO3α and PGC-1α proteins. BHTF of low dose can improve the expressions of FOXO3α and PGC-1α proteins. Increase the blood supply of the brain,thereby protecting the brain tissue of CCCI rats.
