腹主动脉瘤组织中HIF-1α、MMP-2及MMP-9的表达及意义 被引量:4

摘要 目的探讨低氧诱导因子(HIF)-1α、基质金属蛋白酶(MMP)-2及MMP-9在腹主动脉瘤(AAA)组织中的表达及意义。方法采用免疫组化法检测68例AAA及20例正常腹主动脉组织中HIF-1α、MMP-2及MMP-9的表达,并分析三者与临床病理参数的关系。结果 HIF-1α表达于AAA中膜及外膜平滑肌细胞、巨噬细胞的细胞核中,MMP-2表达于AAA中膜及外膜平滑肌细胞的细胞质中,MMP-9表达于AAA中膜及外膜平滑肌细胞、巨噬细胞的细胞质中,三者在正常腹主动脉组织中均不表达。AAA中HIF-1α、MMP-2及MMP-9的阳性率显著高于正常腹主动脉(P<0.001)。AAA中动脉瘤直径≥5.0 cm的患者HIF-1α、MMP-2及MMP-9的表达高于动脉瘤直径<5.0 cm者(P<0.001);AAA中有发生心脑血管意外史患者HIF-1α、MMP-2及MMP-9的表达高于未发生者(P<0.001)。AAA中HIF-1α、MMP-2及MMP-9与年龄、性别、有无吸烟史及高血压无关。同时AAA中HIF-1α、MMP-2及MMP-9表达两两呈正相关。结论 HIF-1α在AAA中高表达,并与MMP-2及MMP-9起协同作用,在AAA的发生发展中起重要作用。
