康力欣胶囊对人脐静脉血管内皮细胞周期和凋亡的影响

Effect of Kanglixin capsules on on cell cycle and apoptosis of human umbilical vein endothelial cells

目的研究抗肿瘤药康力欣胶囊对人脐静脉血管内皮HUVEC细胞增殖、细胞周期和细胞凋亡的影响。方法以康力欣胶囊接种于细胞12、24、36、48 h后,采用四甲基偶氮唑盐(MTT)法测定细胞抑制率,采用流式细胞仪检测细胞周期各时相比率的变化,荧光显微镜观察法检测各组细胞凋亡的形态变化。结果 MTT检测结果显示康力欣胶囊能够明显抑制HUVEC的增殖,其抑制率与药物浓度呈正相关;细胞周期检测结果显示,康力欣胶囊将HUVEC细胞周期阻滞在S期;荧光显微镜观察结果显示,康力欣胶囊作用HUVEC后出现大量凋亡细胞。结论康力欣胶囊对HUVEC有明显的抑制作用,该作用可以通过阻滞细胞周期和诱导细胞凋亡实现。

Objective To observe the effect of the antitumor medicine Kanglixin capsules on cell proliferation, cell cycle and apoptosis of the HUVEC. Methods After inoculated for 12, 24, 36, and 48 hours respectively, the action of Kanglixin capsules on growth inhibiting rate was detected by MTT, the DNA content of cell cycle in each phase was detected by flow cytometry and the morphologic change of cell apoptosis was observed through fluorescence microscope. Results The result of MTT method reveals that the inhibition rate of Kanglixin capsules has a positive correlation with the drug concentration; the flow cytometry result indicates that Kanglixin capsules can block cell cycle in S phase; the fluorescence microscop e observes that Kanglixin capsules could induce apoptosis of HUVEC. Conclusion The inhibition of Kanglixin capsules on HUVEC can be realized by means of blocking cell cycle and induce the cell apoptosis.
