摘要
目的探讨儿童肺炎与TNFa的相关性。方法采用序列特异性引物—聚合酶链反应(SSP/PCR)对265例肺炎患儿和265例随机来院体检的健康儿童TNFa的5个等位基因进行分析,计算各个位点的基因分布频率,并进行卡方检验,肺炎组与对照组之间进行逐个等位基因的比较,计算相对危险性RR值,筛选有意义基因。结果 265例肺炎患儿与265例健康儿童TNFa等位基因卡方检验χ2=23.611,P=0.023<0.05,说明肺炎组和对照组TNFa等位基因基因频率有明显差异,肺炎组与对照组之间进行逐个等位基因的比较,-1031 TT、-1031TC、-1031CC(108+25bp)、-1031CC(360+161bp)、-863CA、-863AA、-857 CT、-857GG、-308 GA、-308 GG、-238GA两组间有显著性差异,其他位点两组间无显著差异。-1031 TT、-1031TC、-1031CC(108+25bp)、-1031CC(360+161bp)、-863CA、-863AA、-857 CT、-857GG、-308 GA、-308 GG基因阳性肺炎的相对危险性RR值分别为1.57、2.64、4.80、2.64、2.91、2.75、2.25、2.92、2.07、2.42。结论 TNFa可能是儿童肺炎的易感基因。
Objective:To investigate the relationship between children with pneumonia and TNFa. Methods:TNFa 5 alleles frequencies Were measured in 265 children with pneumonia and 265 healthy children using polymcerase chain reaction with sequence specific primer,and calculate each allele frequencies and relative dangemusne RR value;and operate chi-sqare test,so that selecting significant genes. Results:chi-sqare test were operated in 265 children with pneumonia and 265 healthy children,χ2=23.611,P=0.0230.05,so we can included that there was significant difference between pneumonia group and normal group in Allele frequency,Every allele is compared,there was significant difference between the two groups in-1031 TT,-1031 TC,-1031 CC(108+25 bp),-1031 CC(360+161 bp),-863 CA,-863 AA,-857 CT,-857 GG,-308 GA,-308 GG,-238 GA,there was no significant difference in other alldes.the RR value of-1031 TT,-1031 TC,-1031 CC(108+25 bp),-1031 CC(360+ 161 bp),-863 CA,-863 AA,-857 CT,-857 GG,-308 GA,-308 GG is 1.57,2.64,4.80,2.64,2.91,2.75,2.25,2.92,2.07,2.42. Conclusions:TNFa possibility were the predisposing genes of children with pneumonia.
