桑叶源γ-氨基丁酸定量分析及富集技术初探 被引量:2

摘要 采用柱前衍生-高效液相色谱技术建立了桑叶中γ-氨基丁酸(GABA)定量分析方法,并研究了厌氧气调、厌氧/好氧交替气调方式对桑叶中GABA富集效果的影响。气调处理能有效提高桑叶中GABA提取量,不同桑叶品种、不同气体种类处理结果存在差异,且交替气调处理相比单纯厌氧处理对GABA富集效果更好。烟台小树桑叶经CO2处理3 h,增氧1 h,交替处理2次得到的桑茶GABA提取量为2.26 mg/g,为空白样品的6.27倍,富集效果最显著。该方法线性关系良好(R2=0.998),线性范围为0.01~0.40 mmol/L,可以应用于桑叶源γ-氨基丁酸定量分析。 The quantitative analysis method of GABA in mulberry leaves was established by pre-col- umn derivatization high performance liquid chromatography (HPLC). The effects of anaerobic or an- aerobic/aerobic alternating atmosphere regulate on GABA enrichment were studied. The results showed that the GABA content in modified mulberry leaves was higher, and the effects were different for diverse mulberry varieties and gas types. And the GABA enrich effect of alternating gas treatment was better than that of simple anaerobic treatment. The most significant treatment was 02 treat for 1 h after CO2 treat for 3 h, and repeat it once, the GABA content increased 6.27 times to 2.26 mg/g. Linear calibration of GABA standard was good (R2 = 0. 998 ). The linear range of the method was 0.01 - 0.40 mmol/L. The method can be applied to quantitative analysis of GABA in mulberry leares.
