摘要 为了研究UGT基因家族在石榴中的作用和表达方式,对石榴UGT基因家族的理化性质、选择压和蛋白基序等进行分析,选择桃、葡萄、桉树、甜橙5个物种的UGT基因家族与石榴UGT基因家族进行比较基因组分析。结果表明,桃有最多的UGT基因(58个);桉树有52个,位列第2;葡萄最少,只有15个;石榴(32个)约是葡萄的2倍;甜橙(39个)和石榴数量相似。选用MEGA软件中的邻接距离矩阵法构建系统进化树,应用自展法进行可信度检测,重复1 000次。进化树表明,UGT基因家族被分成5个亚群。通过选择性压力分析估计UGT基因家族中非同义突变(d_N)与同义突变(d_S)的比值,结果表明d_N/d_S<1,即UGT基因家族在桃、葡萄、石榴、桉树、甜橙5个物种中以纯化选择为主。UGT基因家族的蛋白基序分析结果表明,UGT在低阀值(E=1.1e-2 355)时呈现"HPAVGGFLTHCGWNS"的主题。另外,相对分子质量的平均估计值(-0.095 183 7),表明UGT基因所编码蛋白有亲水性。 In order to study function and expression means of UGT gene family in Punica granatum, physical and chemical properties, selection and motif of UGT gene family in P. granatum were analyzed, and a comparative genomics research on UGT gene families in Prunus persica, Ntis vinifera, Eucalyptus grandis, Citrus sinensis and P granatum was carried out. The results showed that, P persica had the maximum members of UGTgene family (58), E. grandis was the second (52), and V. vinifera had the minimum number (15). The UGTmembers (32) in P granatum was as roughly two times many as which in V. vinifera. Similar results were surveyed on C. sinensis genome, and a total of 39 UGT genes were identified. A phylogenetic tree was constructed by using adjacency distance matrix method in MEGA, reliability was determined by using bootstrap method, and repeating 1 000 times. The phylogenetic tree showed UGT gene family was divided into five subgroups. Ratio of non-synonymous (dN) to synonymous mutations (ds) was estimated by using selective pressure analysis. The results showed that dN/ds 〈 1, that was, UGT gene family predominated purifying selection among the five species of P persica, V. vinifera, P granatum, E. grandis and C. sinensis. The results of motif analysis showed that UGTgene family had a motif "HPAVGGFLTHCGWNS" with a low E-value (1.1e - 235 5). UGTgene encoded a hydrophilic protein, which was inferred based on average relative melocular mass ( -- 0.095 183 7). Keywords: comparative genomics, Punica granatum; UGT gene family
