摘要
[目的]探讨马钱子碱对乳腺癌骨转移相关因子的影响。[方法]建立裸鼠乳腺癌骨转移的模型,并用马钱子碱干预后用逆转录聚合酶链反应法(RT-PCR)检测各组肿瘤组织中骨转移相关因子MMP-2、CXCR4、RANKL、OPG m RNA表达量。[结果]马钱子碱经腹腔给药的半数致死量(Medial lethal dose,LD_(50))为69mg/kg。根据LD50的1/10、1/20和1/40的量,设三个剂量组:高剂量组6.90mg/kg1、中剂量组3.45mg/kg和低剂量组1.73mg/kg,同时与空白对照组比较。马钱子碱各剂量组MMP-2,CXCR4,RANKL m RNA的表达量随着马钱子碱剂量的增加逐渐下降(P<0.05),而OPG m RNA的表达量则随着马钱子碱剂量的增加逐渐增加(P<0.05)。[结论 ]马钱子碱可以抑制乳腺癌骨转移,主要以调节MMP-2、CXCR4、RANKL、OPG等骨转移相关因子的表达为主要手段。
[Objective] To explore the effect of brucine on bone metastasis factors in breast cancer. [Methods] Model of breast cancer bone metastasis in nude mice was made,bone metastasis correlation factors expression of MMP-2,CXCR4,RANKL,OPG m RNA after Brucine intervention were detected by RT-PCR method. [Results] The median lethal dose(LD(50)) of brucine by intraperitoneal administration was 69 mg/kg. According to LD50 and 1/10,1/20 and 1/40,the experiment group was divided into high dose group(6.90 mg/kg),middle dose group(3.45 mg/kg),low dose group(1.73 mg/kg) and control group. The MMP-2,CXCR4,RANKL m RNA expression decreased with Brucine dose increased(P 0.05), While the expression of OPG m RNA increased with Brucine dose increased(P〈0.05). [Conclusion ]Brucine can inhibit bone metastasis of breast cancer,mainly in regulating bone metastasis-associated factors such as MMP-2,CXCR4,RANKL,OPG as the main means of expression.
