脑络欣对大脑中动脉阻塞再灌注模型大鼠海马区细胞层面的影响

Influence of Naoluoxin on the Cellular Level of the Hippocampal Area in the Model Rats with Middle Cerebral Artery Occlusion and Reperfusion

摘要 目的:观察脑络欣通对大脑中动脉阻塞再灌注(Middle cerebral artery occlusion and reperfusion,MCAO-R)大鼠海马区细胞层面的作用机制与效应。方法:将30只大鼠编号称体质量、记录、计算麻醉剂用量后随机分为假手术组6只和手术组24只。手术组大鼠采用线栓法造模,将造模成功的大鼠随机分为模型组和给药组,每组各12只。造模后第1天,予给药组大鼠脑络欣通溶液1 mL·kg^(-1)灌胃,假手术组和模型组予等量生理盐水灌胃,共7 d。制备大鼠脑组织石蜡切片并进行HE染色。结果:假手术组大鼠脑组织缺血面积为0,模型组和给药组脑缺血面积百分比比较,差异具有统计学意义(P<0.05)。模型组细胞排列散乱,锥体细胞数量减少,胶质细胞增生明显;给药组锥体细胞排列比较规则,在多个区域数量明显增多,甚至有迁移现象,偶见胶质细胞。结论:脑络欣通对大脑神经细胞不仅有促进增殖和分化的作用,而且能够促进神经干细胞在增殖分化后迁移,海马CA1、CA3区尤其明显,以替代缺血区损伤及死亡神经细胞,从而改善神经功能,促进脑功能的恢复。 Objective:To observe the mechanism and effect of Naoluoxin on the cellular level of the hippocampal area in the model rats with middle cerebral artery, occlusion and reperfusion ( MCAO-R ). Methods: Totally 30 rats were numbered, weighed body mass, calculated the dosage of anesthetics and randomized into sham operation group (6 rats) and operation group (24 rats). Op- eration group were conducted suture-occluded method for modeling. The rats 'after successful modeling were randomized into model group and administration group, 12 rats in each group. One day after modeling,administration group were given garage of Naoluox- in Solution of lml/kg while sham operation group and model group were given same quantily of normal saline,7 days for each group. The paraffi,1 section of the brain vessel were prepared and given HE staining. Results:The area of tissue ischemia was 0 in sham operation gruup;the difference of the area of tissue ischemia between model group and administration group had statislieal significance ( P 〈 0.05 ). The cell an'angement in model group was scattered,the number of pyramidal cells reduced, and the glial cells fihyperplasia was obvious. The cell arrangement in administration group was regular,the number increase was ohvious,there was even transport phenomenon,and glial ceils occurred oeeasiovrally. Conclusion: Naoluoxin has proliferation and differentiatio,1 action,which can promote the transfer of neural stem cells "after proliferation and differentiation especially CAI and CA3 in hipp- oeampal area to replace the injury of ischemie region and dead nerve cells and improve the neurologic function and recovery of the brain function.
