摘要
目的:探讨补肾滋阴、补肾温阳法对糖尿病性勃起功能障碍(Erectile dysfunction,ED)大鼠雄激素的影响,并探讨不同药物剂量下雄激素变化趋势。方法:将64只雄性大鼠,随机选取8只作为空白对照组,余56只腹腔注射链尿佐菌素(STZ)构建糖尿病性(diabets mellitus,DM)大鼠模型,成模后用阿朴吗啡(APO)筛选DM性ED模型,造模成功后随机分为模型组、补肾滋阴方低剂量组、补肾滋阴方中剂量组、补肾滋阴方高剂量组、补肾温阳方低剂量组、补肾温阳方中剂量组、补肾温阳方高剂量组,每组各8只。参照《药理实验方法学》,计算给药剂量。将补肾滋阴方及补肾温阳方药物加水至112 mL,其中高剂量组灌胃量等于大鼠公斤体质量数×系数2×2 mL;中剂量灌胃量:公斤体质量数×系数1×2mL;低剂量灌胃量:公斤体质量数×系数0.5×2 mL。每日1次灌胃;给药共4周。模型组及对照组给予等容积生理盐水灌胃。药物干预四周后测定血清睾酮(testosterone,T)水平。结果:与空白对照组比较,模型组T水平显著下降,差异具有统计学意义(P<0.05);与模型组比较,补肾滋阴组、补肾温阳组T水平显著升高,差异具有统计学意义(P<0.05);补肾滋阴与补肾温阳两组比较,差异无统计学意义(P>0.05)。与模型组比较,补肾滋阴中剂量组、补肾滋阴低剂量组T水平升高,差异具有统计学意义(P<0.05),补肾滋阴高剂量T水平虽然升高,但是差异无统计学意义(P>0.05)。与模型组比较,补肾温阳中剂量组、补肾温阳高剂量组T水平升高,差异具有统计学意义(P<0.05),低剂量组T水平虽然升高,但是差异无统计学意义(P>0.05)。结论:补肾滋阴法、补肾温阳法均可提高DM性ED大鼠雄激素水平,其中补肾滋阴法中剂量疗效最佳,高剂量疗效反而下降,应避免大剂量使用滋阴药;补肾温阳法随药物浓度增加疗效增加,可适当增加温阳药物剂量。
Objective : To explore the influence of the method of nourishing kidney yin and supplementing kidney yang on the andro- gen of rats with erectile dysfunction (ED) due to diabetes mellitus ( DM), and further explore the variation trend of androgen with different dosage of drugs. Methods :Totally 64 male rats were selected. Among them ,8 male rats were randomly selected as blank control group and the rest of the 56 male rats were injected with streptozotozin (STZ) to make experimental models of DM, then injected with apomorphine (APO) in all of the DM rats to select erectile dysfunction rats models with diabetes. The ED rats were randomly divided into model group, nourishing kidney yin of low dosage group, nourishing kidney yin of medium dosage group, nourishing kidney yin of high dosage group, supplementing kidney yang of low dosage group, supplementing kidney yang of medi- um dosage group and supplementing kidney yang of high dosage group,8 rats in each group. Kidney Yin-Nourishing Prescription and Kidney-Yang Supplementing Prescription were added with water to 112 mL. High dosage group were given gavage volume e- qualing body mass 2 × 2 mL; medium dosage group were given gavage volume equaling body mass 1× 2 mL; low dosage group were given gavage volume equaling body mass 0.5 × 2 mL, once every day and for consecutive 4 weeks of administration. Model group and control group were given same volume of normal saline for gavage. After 4 weeks of chug intervention, the levels of tes- tosterone (T) were determined. Results:Compared with blank control group, the level of T in model group declined obviously, with statistical difference ( P 〈 0.05 ). Compared with model group, the levels of T in nourishing kidney yin group and supplementing kidney yang group increased significantly, with statistical difference ( P 〈 0.05 ). The difference between nourishing kidney yin group and supplementing kidney yang group had no statistical significance ( P 〉 0.05 ). Compared with model group, the levels of T in nourishing kidney yin of medium dosage and high dosage groups increased,with statistical difference (P 〈0.05 ). The level of T in low dosage group increased but there was no significant difference (P 〉 0.05 ). Conclusion:The methods of nourishing kid- ney yin and supplementing kidney yang can enhance the levels of androgen in rats with ED due to DM. The effect of nourishing kidney yin of medium dosage group is the best;high dosage may reduce the efficacy, so the use of high dosage of yin-nourishing drugs should be avoided. The increase of concentration of the drugs in the method of supplementing kidney yang will improve the efficacy, so it is advisable to increase the dosage of yang-warming drugs appropriately.
