摘要 目的以通关藤为原料分离纯化得通关藤多糖组分TCS4,并进行初步结构分析。方法将通关藤多糖原料精制,大孔树脂柱除蛋白和色素,离子交换纤维素DEAE-32、葡聚糖凝胶Sephedex G-50分级分离,聚丙烯酰胺凝胶电泳鉴定纯度,高效凝胶渗透色谱测定相对分子质量,1-苯基-3甲基-5-吡唑啉酮柱前衍生化高效液相色谱、红外光谱、高碘酸氧化、smith降解、核磁共振等方法初步确定结构。结果分离纯化得酸性均一多糖组分TCS4,相对分子质量约7000,单糖组成为甘露糖、鼠李糖、葡萄糖醛酸、半乳糖醛酸、葡萄糖、半乳糖、阿拉伯糖,摩尔比为1∶3.5∶1.9∶6.9∶7.8∶10∶4.7。结论通关藤多糖TCS4为首次提取出的小分子量酸性均一杂多糖,不同产地通关藤多糖组成的比例有一定差异。 OBJECTIVE To isolate and elucidate the chemical structure of a homogeneous polysaccharide TCS4 purified from Marsdeniae tenacsima. METHODS TCS4 was obtained by purification with macroporous resin and ultrafihration from raw polysaccharides. The purities andmolecular weight of TCS4 were identified by High perform- ance gel permeation chromatography. Pre-column derivatization HPLC, IR, periodate oxidation, Smith degradation, NMR were used to determine the structure of TCS4. RESULTS The molecular weight of TCSd was about 7000, and consisted of mannose, rhamnose, glucuronic 1.0:3.5: 1.9:6. 9:7.8: 10:4.7. The major acid, galacturonic acid, glucose, galactose, arabinose with themolar ratio of linkage of TCS4 was or- ( 1-4 ) glucose, glucuronic acid and rhamnose. The branches of TCS4 were arabinose and galactose terminal linked, and small amount of (1-3) galac- tose. CONCLUSION This polysaccharide is a new hetero-polysaccharide from Marsdeniae tenacissima, and its com- position ratio was different with places of produce.
