利用广谱抗病基因Pigm改良水稻稻瘟病抗性 被引量:8

The Improvement of Rice Blast Resistance by Broad-spectrum Gene Pigm

摘要 为改良已选育的高产优质粳稻高代新品系的稻瘟病抗性,以引进的携带有水稻广谱抗稻瘟病基因Pigm的籼稻品种谷梅4号和粳稻材料MP3为供体,利用花粉管通道技术将其基因组DNA导入到SD-2、SD-3、SD-4、SD-5、SD-6、DJ13、ZD15、14PB1和14PB4等高代品系和品种宁粳43号中,利用Pigm基因的显性标记M26205和M80410对受体及获得的导入系进行检测,结果表明:在所有受体中,两个标记均未扩增出条带,说明所有受体本身都不含Pigm基因;在检测的854株导入株系中,利用标记M26205检测总共有328株能够扩增出856 bp,或1 019 bp和856 bp的条带,初步推断这328个株系携带有Pigm基因,平均阳性株率为40.51%;利用标记M80410检测总共有169株能够扩增出517 bp的条带,推断这169个株系可能携带有Pigm基因,平均阳性株率为19.83%。选择两个标记能同时检测出的阳性植株107株作为Pigm基因的导入系,进行进一步的加代培育和辅助选择。 To improve the bla st resistance of high generation japonica rice breeding lines which has high yield and good quality, the indica rice variety Gumei4 and japonica rice materials MP3 carried broad-spectrum blast resistance gene Pigm were imported as donor, and the pollen tube channel technology were used to import the donor's genomicDNA into breeding lines of SD-2, SD-3, SD-4, SD-5, SD-6, DJ13, ZD15, 14 PB1, 14 PB4 and variety Ninggeng43, the dominant marker M26205 and M80410 of the Pigm gene were used to detect the receptor and the introgression lines. The results showed the 2 markers didn't amplified any band in all receptors, that revealed all the receptors didn't carry Pigm gene; In all 854 introgression lines, a total of 328 plants can be amplified to 856 bp, or 1 019 bp and 856 bp band by marker M26205, It can be conclud that the 328 plants carried Pigm gene, the average positive plants rate was 40.51%; A total of 169 plants can be amplified to 517 bp band by marker M80410, It deduced that the 169 plants carried Pigm gene, the average positive plants rate was 19.83%. A total of 107 plants can be detected positive by both markers were selected as the introduction of Pigm gene, and then for further breeding and molecular marker assisted selection.
