摘要
SSR标记在真实性检测、纯度鉴定、以及指纹库构建中起着至关重要的作用。在其筛选过程中会遇到许多具有N+1峰等异常峰型、非特异性峰、多态性低、不适于软件自动分析等非优异SSR引物。为了解并掌握非优异SSR引物表现以及形成原因,本研究以22份初筛材料、27套三联体材料及95份自交系对新型SSR引物进行评估筛选,从中选取有代表性的11对SSR引物,现在已对10种非优异SSR引物表现进行描述,分析形成原因,通过测序对一对引物进行重新设计,并获得较好效果。
SSR markers play an important role in authenticity detection, purity identification and fingerprint library building In the process of SSR primers screening will meet imperfect SSR primers, such as N +1 peak and other abnormal peak, nonspecific peak, low polymorphism and not suitable for automatic analysis software. In order to understand and master the performance and the reasons about imperfect SSR primers, in this study, using22 materials about first screening, 27 sets of mother-father-child trios and 95 inbred lines for evaluate new SSR primers screening and choose 11 pairs representative primer,now in 10 kinds of imperfect SSR primers was described expression and analyzes the formation reasons. Redesigned a pair of primer by sequencing and obtain satisfactory result.
