摘要
本研究以吸附率、解吸率为考察指标,采用静态吸附方法筛选出纯化效果最佳的LSA-21型树脂。考察各种因素对树脂吸附、解吸效果的影响,优化得到动态吸附最佳工艺条件为:上柱液浓度3.12 mg/m L,上样量203 m L,吸附速率3 BV/h,上柱液pH为6,解吸剂乙醇浓度70%,解吸剂用量180 m L。在此工艺条件下,吸附率及解吸率平均值分别为91.83%和91.41%,瓦松干浸膏中总三萜成分纯度从9.36%提高到40.56%,因此该工艺可以有效地纯化瓦松总三萜成分。
In this research the adsorption and desorption ratio of total triterpenoids were used as evaluation indexes, the suitable LSA-21 macroporous resin was screened with the method of static adsorption. And the optimum technological condition was optimizated by investigating the effects of many factors on the adsorption and desorption of the resin, which were original solution concentration of 3.12 mg/m L, the loading quantity of 203 m L,adsorption rate of 3 BV/h, the original solution of 6 pH value, desorbent ethanol concentration of 70% and the eluants volume of 180 m L. In this condition, the adsorption and desorption ratio were 91.83% and 91.41%, respectively. The purity of total triterpenoids in dry extracts could be changed from 9.36% to 40.56%, so the technology was effectively for the purification of total triterpenoid saponins from Orostachys fimbriatus(Turcz) Berger.
