MALDI-TOF MS技术在临床感染猪肠球菌检测中的应用研究

摘要 为评价基质辅助激光解吸电离飞行时间质谱(MALDI-TOF MS)技术在肠球菌的鉴定及同源性分析中的应用,利用10株经16S rRNA确认的肠球菌建立了肠球菌指纹图谱数据库,建库后使用MALDI-TOF MS技术对河南省部分地区临床感染猪分离得到的33株肠球菌野生株进行鉴定并进行了同源性分析。结果表明:在33株肠球菌中,鉴定出粪肠球菌13株、屎肠球菌20株,且可信度分值大于2.300,鉴定符合率为100%。说明MALDI-TOF MS技术可对肠球菌进行快速鉴定和同源性分析,操作快速、简便,准确率高,有利于兽医临床上病原菌的快速诊断。 To investigate the application of Matrix Assisted Laser Desorption Ionization Time of Flight Mass Spectrometry( MALDI-TOF MS) in the identification and homology analysis of Enterococcus,database of mass spectrum map of Enterococcus was created using 10 strains of Enterococcus identified by 16 S rRNA amplification. Then the database of mass spectrum map of Enterococcus was applied to the identification and homology analysis of 33 wild strains of Enterococcus in Henan area. The results showed that during 33 strains of Enterococcus,13 isolates were E.faecalis and 20 isolates were of E. faecium. The confidence score was greater than 2. 300 and Qualification rates were 100%. These indicate that the method of MALDI-TOF MS can be used in the identification and homology analysis of Enterococcus,which is rapid,simple and accurate,and is helpful for the rapid diagnosis of pathogenic bacteria in veterinary clinic.
