急性肺损伤ACE变化及地塞米松的拮抗作用 被引量:2

The changes of ACE in endotoxin-induced acute lung injury and antagonistic effects of dexamethasone

摘要 目的:探讨地塞米松对内毒素(ET)所致急性肺损伤(ALI)中血清血管紧张素转换酶(ACE)活性的影响,为临床ALI的药物选择提供理论依据。方法:将24只3个月龄雄性白兔随机分为生理盐水组(A组),ET组(B组),地塞米松+ET组(C组)。用静脉注射ET 700μg·kg^(-1)方法制作ALI模型,C组在ET注射前给予地塞米松(5 mg·kg^(-1))干预,在实验0、0.5、1、2、4 h行血氧饱和度(Pa O_2/Fi O_2)和血清ACE检测。结果:B组Pa O_2/Fi O_2在0.5 h降至最低为(265.4±70.2)mm Hg,随后逐渐升高,血清ACE活性在0.5 h升高至(68.39±6.29)u·m L^(-1),在1 h达到峰值(80.13±12.97)u·m L^(-1),2 h开始逐渐下降。C组Pa O_2/Fi O_2也呈现下降趋势,血清ACE活性呈现升高趋势,与B组比较具有显著性差异,与A组比较差异无统计学意义。结论:地塞米松可通过对血清ACE活性的抑制,对ET所致ALI产生一定的保护性作用。 Objective:To study the effects of dexamethasone on the changes of serumal angiotensin converting enzyme(ACE)activity in endotoxin(ET)-induced acute lung injury(ALI)and provide references for the choice of the ALI drugs in ckinic.Methods:The 24 rabbits were divided into three groups:saline control group(group A),ET injuried group(group B),dexamethasone+endotoxin group(group C).Rabbit model of ALI was induced by ET administered intravascularly with a dose of ET 700μg·kg-1.The rabbits in group C were injected with a dose of dexamethasone(5 mg·kg-1)before ET.The changes of blood oxygenation index(Pa O2/Fi O2)and serumal ACE activity were observed at 0 h,0.5 h,1 h,2 h,4 h respectively.Results:In group B,Pa O2/Fi O2 decreased significantly at 0.5 h(265.4±70.2 mm Hg),then gradually increased.In group B,serumal ACE activity signifi cantly increased at 0.5 h(68.39±6.29 u·m L-1),reached at peak at 1 h(80.13±12.97 u·m L-1),gradually decreased after 2 h.In group C,the Pa O2/Fi O2 also decreased.The activities of serumal ACE were signifi cantly lower than those in group B.The Pa O2/Fi O2and the activity of serumal ACE were comparable to group A.Conclusion:Dexamethasone can possess some protective effects on ALI by inhibiting serumal ACE activity.
