摘要
采用超声法提取关黄柏皮、叶和关黄柏果中盐酸小檗碱,并用高效液相色谱法测定其不同部位盐酸小檗碱含量,流动相为甲醇-乙腈-磷酸(0.01 mol/L)=(10∶28∶62,V/V),流速为1 m L/min,检测波长为350 nm;进样量为10μL.线性范围为2.20~13.20μg/m L,标准偏差(RSD)小于2.70%,加标回收率为93.21%~99.38%.实验结果表明:关黄柏皮、叶和果中盐酸小檗碱含量分别为:0.484%,0.323%,0.002 1%,关黄柏叶和皮中均含有较高含量的盐酸小檗碱,其中关黄柏皮盐酸小檗碱的含量最高.
Ultrasonic method was used to extract berberine hydrochloride in cortex phellodendri skin,leaves and fruit,high-performance liquid chromatography(HPLC) determination of the contents.The mobile phase of methanol,acetonitrile-phosphate(0.01 mol/L) =(10 ∶ 28 ∶ 62,V/V),the flow rate of 1 m L/min,detection wavelength of 350 nm; The sample quantity is 10μL.The linear range is 2.20 ~ 13.20 μg/m L,the standard deviation(RSD) is less than 2.70%,and the recovery rate of the standard is 93.21% ~ 99.38%.The experimental results show that the berberine hydrochloride in cortex phellodendri skin,leaves and fruit content 0.484%,0.323%and 0.002 1% respectively.Cortex phellodendri skin and leaves are all contain a higher concentration of berberine hydrochloride,cortex phellodendri skin contain the highest content of berberine hydrochloride.
