摘要
为探讨己糖激酶2(HK2)基因与鹅脂肪肝形成的关系,选取30只朗德鹅为研究对象,将其分为填饲组(15只)和对照组(15只),填饲组分填饲7、14和19d3个阶段。采用实时荧光定量PCR技术测定朗德鹅填饲不同阶段HK2在肝脏、胸肌和腹脂中的表达水平,并用葡萄糖、脂肪酸和胰岛素分别处理鹅原代肝细胞,观察这些因子对HK2基因表达的影响。结果表明:各阶段填饲组肝脏中HK2基因的表达量显著高于对照组,胸肌和腹脂则呈下调趋势。此外,在培养的鹅原代肝细胞中,葡萄糖、不饱和脂肪酸(油酸和亚油酸)能使HK2基因表达量显著下调,而胰岛素能显著上调HK2基因表达。HK2基因表达与鹅肥肝形成密切相关,为深入研究HK2基因在鹅肥肝形成过程中的作用奠定了基础。
To explore the relationship between HK2 and liver fatty in goose, 30 Landes geese were divided into over- feeding group (15) and control group (15). The overfeeding group included 3 periods., overfeeding 7, 14 and 19 days. HK2 expression levels at different stages of overfeeding were determined in both normal and fatty liver by qRT-PCR in Landes geese. In addition, primer hepatocytes feom goose were treated with glucose, insulin or fatty acids to determine how HK2 expression was regulated during fatty liver formation. Data indicated that the HK2 expression in the livers of overfed geese was significantly higher (P〈0.05) than that in the control at every stage, while HK2 expression was in a downward trend in the abdominal adipose tissues and breast muscles. Moreover, compared to the control, HK2 expres- sion was significantly decreased in primary hepatocytes from goose treated with glucose and unsaturated fatty acid (oleate& linoleate) (P〈0.05), but was increased in primary hepatocytes from goose treated with insulin (P〈0.05). The results suggested that HK2 expression is closely related to the development of goose fatty liver, which will provide a basis for further studying the role of HK2 gene in the formation of goose fatty liver.
