摘要
提出了一种新型免疫磁珠细胞分离方法,通过让装有细胞与磁珠悬浊液的试管在环形磁铁中旋转的方式,使磁珠在离心力的作用下向管壁的方向运动,由于越靠近环形磁铁管壁的地方磁场强度越高,磁珠就越容易被吸附.通过对比,旋转分离方法具有更高的分离效率.与此同时,基于该分离方法研制了一款细胞分离仪,将取枪头、脱枪头、移液、旋转试管等操作自动化,更方便临床使用.
Immunomagnetic beads cell separation method is a common method of cell separation. Cell surface antigen binds with specific monoclonal antibody connected with immunomagnetic beads. When magnetic field is applied, cells coupled to magnetic beads are absorbed and retained in the magnetic field.Based on the traditional standing separation method, a new immunomagnetic beads cell separation method was introduced. By rotating tubes containing cells and immunomagnetic beads in the ring magnet, the immunomagnetic beads moves in the direction towards the wall under the effect of centrifugal force. As the distance to the ring magnet becomes closer, the magnetic field strength becomes stronger and the more cells is adsorbed to the tube wall. By contrast with the traditional static separating methods, rotary separation method is more efficient. Based on the rotary method, we developed a cell separation instrument for ease of clinical use by automatically loading tip, removing tip, pipetting, rotating tubes, etc.
